Objective:Electroacupuncture at jiaji points(ex-b2)has been used in ALS patients clinically in China.Data shows that the motor funtion of ALS patient improved significantly by this therapy.To investigate the mechnism of the treatment effects,include onset,survival and motor function,etal.,a control experiment was performanted to test the role of p38MAPK and NF-H in ALS,and find whether electroacupuncture at jiaji points(ex-b2)regulate their expression.Methods:ALS SOD1 G93A transgenic mice were treated by electroacupuncture at jiaji points(ex-b2)with the electrial stimulation at the frequency of 2HZ from the age of d60,20min/time,2 times/week for 4 weeks.The treatment effet was compared with the control group disposed by binding,seizing and nipping the mouse hair at the jiaji points(ex-b2)at the same time.In this study,we proceeded Tail Suspension Test,Turn-club Test to investigated the onset of the disease,survival time of the mice and motor function during the process of the disease,2 times/week from the age of d60 to the end event occurrence.We also tested the expression of p38,p-p38,NF-H,p-NF-H at the veutro horn of the lumbar spinal cord by immunohistochemical staining,and the Level of p38 and p-p38 protein of the lumbar spinal cord by immunoblotting(WB)at the age of day125.All the data obtained through aforementioned procedure was analyzed by the using of SPSS 17.0,gaussian distribution was test first for every group.Data which is in accordance with normal distribution proceeded with independent-samples t test subsequently,and data that is not conform to the normal distribution were going on with rank sum test.Results of the analysis presented as mean value ± standard deviation;Kaplan-Meier Survival analysis was proceeded to analyze the survival datas.We setted p<0.05 stands for the difference has significant meaning.Results:1.Analysis of onset and survival time:EA group shows the delay of the onset of the disease for 9 days compared with the control group,the difference has significant meaning(p<0.01),and the EA group shows a prolong of the survival time of ALS mice for 7 days compared with the control group,the difference has significant meaning(p<0.01).2.Analysis of Weight:Data shows that mice weight of EA group was higher than that of the control group from the age of week21,difference has significant meaning(p<0.05).3.Analysis of Turn-club Test:residence time has declined since the mice age of week16 in control group,and since the mice age of week 17 in EA group.residence time of EA group is significantly longer than that of the control group from the age of weekl6,difference has significant meaning(p<0.05).4 Analysis of IHC:p-p38,p38 and the ratio of p-p38/p38 turned down significantly in the EA group compared with the control group at the age of D125,difference has significant meaning(p<0.05);p-NF-H?NF-H and the ratio of p-NF-H/NF-H turned down significantly in the EA group compared with that in the control group at the age of D125,difference has significant meaning(p<0.05).5.Analysis of western-blot:p-p38 and the ratio of p-p38/p38 turned down significantly in the EA group compared with that in the control group at the age of D125,difference has significant meaning(p<0.05).Conclusion:1.Electroacupuncture at jiaji points(ex-b2)delays the onset of the disease and prolong the survival time of the ALS SVD1G93A transgenic mice.2.Electroacupuncture at jiaji points(ex-b2)improve motor function of the ALS SOD1G93A transgenic mice.3.Electroacupuncture at jiaji points(ex-b2)achieves the treatment effect by downregulating the phosphorylation of p-38 and NF-H.
