The Experimental Study Of Baihui-qubin Acupuncture On Effects Of P-jak?p-stat3?EGFR?GFAP After Acute Intracerebral Hemorrhage In Rats

Objective:To observe the changes of brain tissue morphology in each group under light microscope and the neurological function score and the expression of p-JAK,p-STAT3,EGFR,GFAP protein in rats by acupuncturing scalp,Objective to explore the effects of the possible mechanism of this therapy in the treatment of cerebral hemorrhageMethod:The156 healthy Wistar male rats were selected.(78rats were examined by using light microscope and IHC;78 rats were tested by Western blotting)They were randomly allocated into three groups:blank group.,model group6h,model group1d,model group3d,model group7d,acupuncture group6h,acupuncture group1d,acupuncture group 3d,acupuncture group 7d,AG490 group 6h,AG490 group 1d,AG490 group 3d,AG490 group 7d,12rats in each group.The rats of model group are adopted stereotactic operation in neurosurgery to set up the rat models of intracerebral hemorrhage with reference to the methords which are reported by Rosenberg;The rats of acupuncture group are conducted by using the acupuncture of BaiHui-QuBin and observe the changes of brain tissue morphology in each group under light microscope;The protein expression of p-JAK in the brain tissue of rats in each group were tested by Western Blotting;The expression of p-STAT3?EGFR ?GFAP immunohistochemical method were checked.Results:1 The Result of the neurological deficit scoresAcupuncture can improve the neurological deficit level,and compared with the model group,there were significant differences(P<0.05).2 The Observable Result of Light MicroscopeThe brain tissue arround hematoma in model group became necrotic,dropsical,inflammatory cell infiltration,the shrinkage of nerve cell nucleus,bubbled,the expansion of capillary vessel and hyperemia,the symptom of which became the most serious appeared during the third day.The brain tissue of acupuncture group gradually recovered and the degree of dropsy is light.Moverove,inflammatory cells decreased.3 The Test Result of Western BlotEvery group had protein expressions of p-JAK in different time phase points.The blank group had few expressions.he model group increased expression of p-JAK positive cells when 6h,got peak at 3d,and keep expression till 7d days.Pinprick group and the model group were statistically significant(P<0.05).4The Result of P-STAT Immunohistochemical TestEvery group had positive cell expressions of P-STAT in different time phase points.The blank group had few expressions.The amount of expressions increased obviously after six hours in model group,acupuncture group and AG490 group.P-STAT reached its peak in model group after three days;Compared with model group,the positive cell expression level of P-STAT in acupuncture group decreased obviously(P<0.05).5The Result of EGFR Immunohistochemical Testthe model group 6h EGFR positive cells appeared to increase;got peak at 3d,and keep expression till 7d.Pinprick group compared with model group,were statistically significant at 3d,7d.(P<0.01).6The Result of GFAP Immunohistochemical TestThe results of GFAP immunohistochemistry:model group,GFAP expression appeared to increase at 6h,reached its peak at 3d,keep expression till 7d.Pinprick group compared with model group,were statistically significant at 3d.(P<0.01).Conclusion:1.The treatment of the acupuncture of "Baihui" through“Qubin”can improve neurologic impairment level in the intracerebral hemorrhage,improved neurologic impairment symptoms.penetration2.Acupuncture treatment of“Baihui”penetration“Qubin”can improve nerve cell injury after intracerebral hemorrhage,improve the degree of brain edema on brain and protect neurons.3 Acupuncture treatment of“Baihui”penetration“Qubin”can inhibiting the influence of EGFR?p-JAK?p-STAT and GFAP during the acute period of intracerebral hemorrhage.,so to achieve the function of nerve protection.
