Objective:Breast cancer(BC)is the most common malignancy in women and affects millions of women worldwide.It is essential to understand the mechanisms that promote breast cancer progression.There is evidence that aquaporin(AQPs)are involved in cell proliferation,migration,and peripheral angiogenesis,suggesting that they play an important role in tumorigenesis and growth.This article will study the expression of AQP4 in human breast cancer cells.And further explore the mechanism of downregulation of AQP4 inhibits tumor cell proliferation,migration and invasion.Methods:Breast cancer cells were screened and cultured.The expression of AQP4 was detected by Western Blot assay.The level of AQP4 was down-regulated by siRNA transfection.RNA was extracted,Relevant datas were collected by RT-PCR,Western Blot,cell proliferation assay,scratch test,and Transwell invasion assay,and further will be analysised.Possible signal pathways were discussed and tested to verify,according to the results.Results:(1)AQP4 and E-Cadherin were expressed in human breast cancer cell lines(MDA-MB-231,T47D,MCF-7 and ER-ZR-70).The expression of AQP4 was highest in T47D cells,There is no significant difference in E-Cadherin expression.(2)AQP4 mRNA and protein expression was significantly reduced in cells transfected with siRNA(3)Down-regulation of AQP4 expression decreased T47D cell and MCF-7 cell proliferation,migration,and invasion(4)Down-regulation of AQP4 expression caused increased E-Cadherin expression and inhibited the migration and invasion of breast cancer cells.(5)Down-regulation of AQP4 may reduce the proliferation of breast cancer cells through the MAPK/ERK pathway.Conclusions:(1)AQP4 was expressed in human breast cancer cell lines.(2)Regulation of AQP4 can induce the change of E-Cadherin expression and mediate the proliferation,migration,and invasion of breast cancer cells via MAPK/ERK pathway.
