Objective:The studies of imprinted genes in pregnancy-related diseases are limited,and lack of reliable research evidence.The expression of imprinted genes IFG2(insulin-like growth factor 2)and PHLDA2 CPleckstrin homology-like domain,family A,member 2)in the middle of pregnancy or in preeclampsia has not elucidated.In order to study the expression of IFG2 and PHLDA2 in the middle of pregnancy or preeclampsia,and investigated the expression of IGF2 and PHLDA2 in serum of mid-pregnancy and preeclampsia,whether there is a correlation of PHLDA2 and IFG2 between mid-pregnancy and preeclampsia,exploring the early prediction of mid-pregnancy indicators to preeclampsia,and providing clinical reference or the early intervention of pre-eclampsia.Methods:A total of 40 normal pregnant women and 38 pregnant women with preeclampsia were enrolled in this study from January 2015 to December 2016 in the author's hospital.The pregnant women were divided into three groups by disease severity:normal pregnancy group(n=40),mild preeclampsia group(n=19),and severe preeclampsia group(n=19).The general information of three groups' were collected,the serum of samples(15-20 weeks)and fresh placental tissue after delivery were collected.Levels of serum IGF2 and PHLDA2 in mid-pregnancy were measured by enzyme-linked immunosorbent assay(ELISA),and levels of placental tissue IGF2 and PHLDA2 in late-pregnancy were detected by histochemical staining.The expressions of IGF2 and PHLDA2 mRNA were determined by RT-PCR,and the protein expression of IGF2 and PHLDA2 was detected by Western Blot.The data was analyzed and counted,different groups were analyzed using one-way analysis of variance(ANOVA)and post hoc Fisher 's least significant difference(LSD)test,the correlation analysis were analyzed using Pearson's correlation analysis method,the criterion for significance was set at P<0.05.Results:1.Expression of IGF2 and PHLDA2 in the Placenta TissueCompared with normal pregnancy group,the expression of IGF2 and PHLDA2 RNA and protein in mild preeclampsia group decreased,the difference was statistically significant(P<0.05).Compared with normal pregnancy group,IGF2 and PHLDA2 RNA and protein in severe preeclampsia group decreased,the difference was statistically significant(P<0.01).The expression of IGF2 and PHLDA2 RNA in severe preeclampsia group was significantly lower than that in mild preeclampsia group(P<0.05).Pearson correlation analysis showed that IGF2 expression in placental tissue was positively correlated with PHLDA2 expression(P<0.05).2.Analysis of serum IGF2 and PHLDA2 expression in mid-pregnancyThe expression of IGF2 and PHLDA2 protein in mild preeclampsia group were significantly lower than those in normal pregnancy group(P<0.05).IGF2 and PHLDA2 protein in severe preeclampsia group decreased,the difference was statistically significant(P<0.01).Compared with the mild preeclampsia group,the expression of IGF2 and PHLDA2 protein in severe preeclampsia group decreased,the difference was statistically significant(P<0.05).Pearson correlation analysis showed that serum IGF2 expression and PHLDA2 expression were positively correlated in mid-pregnancy(P<0.05).3 The correlation analysis expression of IGF2 and PHLDA2 in serum and the expression of IGF2 and PHLDA2 in the placentaPearson correlation analysis showed that the expression of serum IGF2 protein in mid-pregnancy was positively correlated with the expression of IGF2 mRNA in the placenta of late-pregnancy(P<0.05).The expression of serum PHLDA2 protein in the mid-pregnancy was positively correlated with the expression of PHLDA2 RNA in the placenta of late-pregnancy(P<0.05).The expression of serum IGF2 protein in the mid-pregnancy was positively correlated with the expression of IGF2 protein in the placenta of late-pregnancy(P<0.05).The expression of serum PHLDA2 protein in the mid-pregnancy was positively correlated with the expression of PHLDA2 protein in the placenta of late-pregnancy(P<0.05).Conclusion:The expression of IGF2 and PHLDA2 in pregnant women with preeclampsia in the third trimester showed a decrease in in the second trimester.The decrease of IGF2 and PHLDA2 may be a high risk factor for preeclampsia in the third trimester.IGF2 and PHLDA2 are closely related to preeclampsia.IGF2 and PHLDA2 may have predictive values for the development of pre-eclampsia...
