?Objective? The effects of Banxia Xiexin decoction on the repair of pancreatic islet injury in DGP rats based on the interaction between intestinal flora and host,observed intestinal flora and intestinal mucosal barrier function of rats,and the experimental basis was provided for widening the application range of Banxia Xiexin decoction and regulating intestinal flora to improve glucose metabolism by compound Chinese medicine.?Method? 120 SPF SD male rats,weighing 200±20 g were randomly divided into normal control group(NG)with 10 rats and DGP model group with 110 rats.After 15 days of feeding with high fat and sugar diet,40 mg/kg STZ was injected into abdominal cavity rapidly after 12 hours of fasting.after 72 hours,the rats were given ordinary granular feed.fasting blood glucose was 16.7~30.0 mmol /l by tail cutting method Metformin group(DGP-met,1.80g/kg);Banxia Xiexin decoction high dose group(DGP-HD,10.2g/kg);Banxia Xiexin decoction middle dose group(DGP-MD,5.1g/kg);Banxia Xiexin decoction low dose group(DGP-LD,2.55g/kg),the other groups were given the same volume of saline,continuous perfusion for four weeks.On the day before sampling,rat feces samples were taken by forced method,intestinal flora structure was detected by 16 sdna method,SCF as was detected by liquid chromatography,and gastric residual intestinal motility,d-xylose content,intestinal barrier and gene expression of islet injury repair related protein were determined after sampling.?Results?(1)compared with NG,STZ-induced DGP rats significantly decreased body weight,intestinal propulsion,fasting blood glucose,gastric residue,serum d-xylose levels were significantly increased(p<0.01).Compared with DGP,Banxia Xiexin decoction can increase body weight and intestinal propulsion,reduce fasting blood glucose,gastric residue and serum d-xylose level(p<0.01).(2)16 sDNA results showed that DGP group decreased the proportion of firmicutes,increased the proportion of bacteroids etes,increased the proportion of bacteroids ia,decreased the proportion of bacteroids and clostridia at the level of phylum,increased the proportion of bacteroids ale,decreased the proportion of lactobacillus and clostriales at the level of phylum,and decreased the proportion of bacteroids at the level of family(3)Compared with NG,the level of SCF as in DGP was significantly decreased,and Banxia Xiexin decoction promoted the production of SCF as significant(p < 0.01).(4)Compared with NG,DGP rats showed obvious damage to intestinal morphology and structure,increased pathogenic bacteria and decreased probiotics(p < 0.01).the levels of ph,muc3,?-defen sin,endotoxin,IgG,IL-6,IL-8,TNF-?,cd8 a increased significantly,and muc2,slga,IL-10 decreased significantly(p < 0.01).Compared with DGP,Banxia Xiexin decoction can improve intestinal microecological environment,reduce pH value,protect mucosal tissue integrity,promote probiotic proliferation,reduce pathogenic bacteria level,regulate intestinal muc-2,muc-3,?-defensin level,significantly reduce endotoxin production(p<0.01),regulate the expression of IgG,slga and cd8 a,reduce inflammatory factors,up-regulate anti-inflammatory factor level(p<0.01)(5)Compared with NG,the levels of IL-10,LPS increased,p-Akt decreased and ERK-1 gene protein increased in DGP rats.compared with DGP,the levels of NGF,PSP / reg gene protein and IGF-1 were significantly inhibited and increased in DGP rats.Compared with DGP,Banxia Xiexin decoction could significantly up-regulate anti-inflammatory factor level,up-regulate p-Akt,down-regulate ERK-1 gene protein expression and reduce LPS production(p<0.01),and had no significant effect on pancreatic NGF.it could regulate PSP/reg gene and protein expression and reduce IGF-1 expression related to islet cell proliferation and differentiation(p<0.01).?Conclusion?(1)DGP can cause hyperglycemia,gastrointestinal motility disorders,intestinal flora disorders,increase serum endotoxin,reduce intestinal flora diversity damage intestinal mucosal mechanical barrier,chemical barrier,biological barrier,immune barrier.(3)Banxia Xiexin decoction can improve gastrointestinal motility disorder and reduce intestinal mucosal permeability in DGP rats;(4)Banxia Xiexin decoction can increase the level of anti-inflammatory factors,reduce the damage of LPS to the liver,has no obvious effect on pancreatic NGF,can regulate the expression of reg-1 gene and protein,regulate the expression of IGF-1 related to islet cell proliferation and differentiation,promote the repair of islet injury,and protect islet function.(5)Banxia Xiexin decoction can improve the diversity of intestinal flora,regulate the ratio of bacteroides to sclerenchyma,increase SCF as,promote probiotic production,inhibit harmful flora and reduce serum endotoxin production;(6)Banxia Xiexin decoction can promote the repair of intestinal tissue injury in DGP rats and protect the integrity of mucosal tissue;Improving intestinal microecological environment and regulating intestinal microecological disturbance caused by DGP;Reduce intestinal flora damage to intestinal mucosal barrier and regulate intestinal mucosal immune response;...
