ObjectivesLiver has tolerant effect,which mediates liver immune tolerance to self and foreign antigen.The 'liver tolerance effect' has been attributed to specialized resident cells and inhibitory cell surface ligands for T cell activation,however,none of studies investigated the role of liver microenvironment on liver immune tolerance while exclude inference of liver resident cells.Decellularized liver scaffold,maintaining the three dimensional structure and extracellular matrix of liver,precisely mimics the liver microenvironment.We hypothesized that decellularized liver scaffold play the major in liver tolerance effect,adopted the liver tolerance effect,and can be potentially used in transplantation as an immunosuppressive material.MethodsMouse liver decellularization was performed.H&E histological analysis and immunofluorescence was used to assess cellulr or nucleic residules.Scanning electrone microscope was used to detect 3D microstructure of the liver scaffold.Flowcytometry analysis was performed to detect the T cell proliferation while elisa detects of cytokines expression in culture supernatant.In vivo islet transplantation was used to determine if decellularized liver scaffold can be further used as an immune suppressive material.ResultsWe found that decellularized liver scaffold,histological confirmed cellular clearance,removes nucleic and cytoskeletal components and widespread preserves extracellular molecules.This scaffold can inhibit T cell activation in vitro by inducing Tcell apoptosis,also,islets transplantation with decellularized liver scaffold improved islet survival compared to control groups.Conclusion.We conclude that decellularized liver scaffold preferentially induced T cell apoptosis and played a crucial role in the tolerogenic nature of the liver.Also,decellualrized liver scaffolds can be utilized as immune-suppressive materials for transplantation.ConclusionsDecellularized liver scaffold preferentially induced T cell anergy via T cell apoptosis and it plays a crucial role in liver tolerant effect.Decellularized liver scaffold co transplantation with islet improved islet survival,can be used as an immunesuppressive material in the induction of immune tolerance in other cell or oragntransplantation.Further research on the specific constituent or protein that led to liver tolerance effect needs to be explored...
