Objective:Analysis of the possible correlation between trace elements,circRNA and papillary thyroid carcinoma,and provide assistance for the diagnosis and treatment of patients with papillary thyroid carcinoma in clinical work.Method From December 2017to October 2018 in xinjiang medical university first affiliated hospital of thyroid surgery in hospital 200 cases of patients with nodular goiter surgery,200 patients with thyroid papillary carcinoma,and in the same period in the first affiliated hospital of xinjiang medical university medical center in healthy check-up crowd of 200 cases were tested for thyroid autoantibodies,serum selenium,zinc,copper and urinary iodine levels.From November 2018 to December 2018,we collected the patients with nodular goiter,thyroid micropapillarycarcinoma?withoutcervicallymphnodemetastasis?,thyroid micropapillary carcinoma with cervical lymph node metastasis 13 respectively in thyroid surgery department of xinjiang medical university first affiliated hospital.The patients were collected 5 ml of preoperative whole blood,for serum circRNA microarray detection,clear circRNA differences between three groups expression spectrum,three cases were selected for RT-PCR verification,and the levels of urinary iodine,blood selenium,blood zinc and blood copper were measured.Seven cases were selected for expanded sample verification.Through the bioinformatics prediction and circRNA high matching values of microRNAs binding sites,Go and KEGG pathway analyses were conducted on differentially expressed circRNA.Results:?1?The differences of TT3,TT4,TSH and TgAb in thyroid papillary carcinoma,nodular goiter and healthy examination group were statistically significant.The differences of urinary iodine,blood selenium,blood copper and blood zinc levels in the three groups were different.All of them were statistically significant.Multivariate logistic regression analysis showed that gender female,high urinary iodine level,low blood selenium level and low serum TT3 level were independent risk factors for thyroid cancer.?2?CircRNA microarray detection was performed in serum of patients with nodular goiter group and papillary thyroid microcarcinoma group,and 14up-regulated and 6 down-regulated circRNA.RT-PCR was performed on circRNA003416,circRNA001359,circRNA104982,circRNA104755,circRNA043614 and circRNA103944,and the results were consistent with those of the chip.Go analysis and KEGG Pathway analysis showed that differentially expressed circRNA mainly by Olfactory transduction pathways to participate in the occurrence of thyroid papillary carcinoma.?3?CircRNA microarray detection was performed in serum of patients with papillary thyroid microcarcinoma without cervical lymph node metastasis and papillary thyroid microcarcinoma with cervical lymph node metastasis,and 400 upregulated and 290down-regulated differentially expressed circRNAs were screened.RT-PCR was performed on circRNA000121,circRNA051239,circRNA001059,circRNA102116,circRNA000466,circRNA001729,circRNA404686,circRNA004183,circRNA102051 and circRNA405571,and the results were consistent with those of the microarray.Go analysis and KEGG Pathway analysis showed that differentially expressed circRNA mainly by other types of o-glycan blosynthesis,salivary secretion,Enolocytosis and other pathways to participate in the occurrence of cervical lymph node metastasis of papillary thyroid microcarcinoma.Conclusion:?1?The nutritional status of human trace elements may affect the incidence of thyroid cancer in the population.Gender female,iodine-rich nutritional status,and lack of selenium nutrition and low serum TT3 level may promote the occurrence and development of papillary thyroid carcinoma.?2?Serum hascircRNA001359 is expected to be a biomarker for the preoperative identification of benign and malignant thyroid nodules in women.?3?Serum hsacircRNA404686 is expected to be a biological marker for the preoperative diagnosis of cervical lymph node metastasis in female with PTMC.
