Objective: To explore the mechanism of Jinshui Liujun Jian in treating airway mucus hypersecretion by observing its effects on COPD“yin deficiency and phlegm retention”rats and A549 mucus hypersecretion cell model.Method: Experiment 1: The model was established by passive smoking+LPS +dexamethasone.Pathological morphology and pulmonary function were used to evaluate COPD.serum c AMP,c GMP,ACTH,CORT and Na+-K+-ATPase were used to evaluate Yin deficiency,and goblet cells and MUC5 AC were used to evaluate phlegm retention.Experiment 2: Azithromycin and Jinshui Liujun Jian at low,medium and high doses were administered on model made in the last part.goblet cells and mucins were detected by PAS staining and MUC5 AC and AQP5 in tissues were detected by RT-PCR and Western blot.Experiments 3: A549 mucus hypersecretion cells model was induced by HNE.CCK8,q RT-PCR,Western blot,ELISA method were applied to verify cell viability,m RNA and protein expression of MUC5 AC and AQP5 in cell and supernatant.Result: Experiment 1: in model group,pathology showed chronic inflammation,emphysema,fibrosis.serum CORT,c AMP,c GMP and Na+-K+-ATPase were decreased(P<0.05),goblet cells was increased(P<0.05),and MUC5 AC was decreased significantly(P<0.01).lung resistance and compliance showed no difference compared with blank group.Compared with model group,CORT,c AMP,c GMP,Na+-K+-ATPase,goblet cells and MUC5 AC were increased in Yangyin group(P<0.05),ACTH,CORT,c AMP,c GMP,Na+K+-ATPase,goblet cells and MUC5 AC in Huatan group showed no differences.Experiment 2: Goblet cells and MUC5 AC were increased(P<0.05),and AQP5 m RNA was down-regulated(P<0.01)in the model group.Compared with model group,MUC5 ACm RNA was down-regulated in medium and high dose groups(P<0.05),and AQP5 m RNA was significantly up-regulated in the medium dose group(P<0.01),AQP5 in the remaining groups were lower than that in the normal group(P<0.05).Experiment 3: MUC5 ACm RNA and protein were significantly increased(P<0.05),AQP5 m RNA and protein were decreased(P<0.05)in model group.Compared with model group,MUC5 AC m RNA in Jinshui Liujun Jian group and Yangyin group was decreased(P<0.05),and AQP5 m RNA in cell and supernatant were increased significantly(P<0.05).protein MUC5 AC in Jinshui Liujun Jian group and Huatan group were decreased(P<0.05),and protein AQP5 in Huatan group was increased(P<0.05).Conclusion: 1.The model established by passive smoking+LPS+dexamethasone was a phlegm retention combined with kidney-yin-yang deficiency model.2.keeping the balance of mucin/water salt ratio may be one of the mechanisms of Jinshui Liujun Jian for treatment of COPD airway mucus high secretion.3.Yin-nourishing effect of Jinshui liujun Jian is related to regulating glucocorticoid-induced adrenocortical depression and cyclic nucleotide metabolism disorder.
