Medulloblastoma is the most common malignant brain tumor in children.Around thirty percent of medulloblastoma patients are diagnosed with metastasis,which often result in poor prognosis.Unfortunately,the metastasis mechanism of medulloblastoma at molecular level remains largely unknown.In this study,we employed the recently developed deep proteome approach to quantitatively profile the expression of around 10,000 proteins from two isogenic cell lines,CHLA-01-MED and CHLA-01R-MED,derived from the primary and metastatic tumor of the same patient diagnosed with medulloblastoma.With the statistic analysis,we identified around 1900 significantly alternated proteins between the primary and metastatic cell lines including the known factors,such as PLGF,LHX1,PROM1,as well as negative regulator SPARC.Further immunohistochemistry analysis of the clinical medulloblastoma samples implied the YAP1 as a potential key factor contributing to the metastasis of medulloblastoma.Taking together,our data broadly defines the metastasis-relevant regulated proteome and provided a precious resource for further investigating the potential mechanism of medulloblastoma metastasis.
