Objective In this study,we firstly would like to focus on papillary thyroid carcinoma and intend to investigate the influential factors of ultrasound elastography(UE).Secondly,we want to investigate what the role of TGF-?1 play in the hardness of thyroid nodules.And finally we would like to investigate what kind of biological effect of GLAG-66,a type of papillary thyroid carcinoma cell will have when inspired by TGF-?1 and its inhibitor.Material and method We retrospectively reviewed the patients underwent thyroid resection surgery for papillary thyroid carcinoma(PTC)in our hospital whose conventional ultrasonography and UE were performed by one doctor.The collagen fibres were stained as pink in the H&E slide and the positive area percent will be calculated by Image J.And then,we would like to investigate different types of thyroid nodules.In the second part,we restrospectively analyzed 102 nodules with different types.Masson staining and immunhistochemical stained were performed to characterize collagen fibres and TGF-?1.The results were calculated by Image pro plus.In the third part,we use a thyroid papillary cancer cell line,GLAG-66.We catagorized the cell in to 3 different group with different cultrued condition and investigated the effect of 3 different groups.All the datas were analyzed by SPSS for Windows,version 19;Chicago,IL.Results In papillary thyroid carcinoma,SR was positively correlated with collagen fibres(r=0.754,p=0.000).If there were insterstitial calcification,SR would elevated dramatically.However,collagen will influence SR more(0.684 vs 0.691),thesize,position,co-existed with HT,multifocality and lymph nodes metastasis had no relation with SR(p>0.05).In different types of thyroid nodules,the expression of collagens and TGF-?1 were positively correlated with SR(r=0.839 and 0.855,p=0.000).In papillaty thyroid cancer,collagen and TGF-?1 expression were positively corrlated with cervical lymph nodes metastasis(0.011 and 0.016),but there was no relation with SR(p=0.287).TGF-?1 aslo had positive effect on invasive of thyroid cell line which could be explained by promoting EMT.Conclusion SR was positively correlated with collagens and calcification of thyroid nodules but the effect of collagen was bigger than calcification.Also similar trend could be observed in different types of thyroid nodules.TGF-?1 and collagen fibre expression were positively correlated with cervical lymph nodes metastasis and TGF-?1 could also enhanced the ability of invasion of GLAG 66,which could be explanied by promoting EMT.
