Background Osteoarthritis(OA)is a common chronic joint disease.The disease has a higher incidence with increasing age and is more common in women.There are many factors that cause osteoarthritis,including heredity,immunity,region,diet,and economy.With the development of science and technology,new technologies and new methods are applied to the treatment of osteoarthritis,but these methods have limitations,mainly in the late stage of osteoarthritis.Osteoarthritis is currently unable to cure,and there is no effective treatment after the onset of the disease,which has a serious impact on patients,their families and society.In recent years,how to effectively prevent and treat osteoarthritis has become an urgent problem to be solved.Long non-coding RNA(LncRNA)is a non-coding RNA of more than 200 nucleotides in length and has a role in a variety of biological processes and diseases,including compensatory effects,epigenetic regulation,cell cycle regulation and regulation of cell differentiation and various diseases including cancer,cardiovascular disease,neurodegenerative diseases,and mental diseases have played an important role.Studies have shown that long non-coding RNA plays an important role in osteoarthritis.Change the expression level of LncRNAs can affect the formation of cartilage and osteogenesis,thereby affecting the homeostasis of cartilage and osteogenesis,and then directly or indirectly participate in bone.The development of arthritis,studies have shown that LncRNAs may help predict the development and diagnosis of osteoarthritis and may help identify potential candidate targets for the treatment of osteoarthritis.However,in damaged cartilage,the expression lineage of LncRNAs is still unknown,and its specific role remains to be studied.Purpose To study and confirm if there are differential expression of LncRNAs and m RNAs between human osteoarthritis and normal cartilage cells,and to explore their potential biological functions.MethodsWe used LncRNAs gene chip screening to obtain gene expression profiles in patients with osteoarthritis and normal population by bioinformatics.By analyzing the gene expression profiles and obtaining some differentially expressed LncRNAs and m RNAs,we classified the differentially expressed LncRNAs and found that most of them belonged to intergenic LncRNA and antisense LncRNA.Analysis of chromosomal distribution revealed that differentially expressed LncRNAs and m RNAs were widely distributed on all chromosomes,including the X and Y chromosomes.To investigate the interaction between LncRNAs and m RNAs,we selected a correlation of 1000 pairs of LncRNAs and m RNAs to analyze the Cytoscape software and found that multiple LncRNAs can simultaneously interact with the same mRNA,as well as antisense.Then using bioinformatics(including GO analysis,Pathway analysis)analysis found that these differentially expressed LncRNAs and m RNAs are closely related to cartilage formation and degradation,such as extracellular matrix formation,ECM receptor interaction,protein degradation and absorption.Finally,we validated differentially expressed LncRNAs and m RNAs(3 expression rises and 3 expression declines)by qRT-PCR in more patient and normal samples.ENPP1 is a type 2 glycoprotein that regulates the metabolism of bone and cartilage tissue by making pyrophosphate to inhibit bone formation.In this paper,Western blot was used to detect the expression of genes related to extracellular matrix by knocking down and over-expressing ENPP1 in primary chondrocytes,thus confirming some functional m RNAs such as COL21 A,ACAN,and ADAMTS5 in early osteoarthritis research.And MMP13.Results Through the above research,we first expressed the expression profile of LncRNAs in osteoarthritis by a new biological means,gene chip technology,and found that these differentially expressed LncRNAs are involved in the biological processes associated with osteoarthritis.A certain role in the signaling pathway provides new insights into the in-depth study of the pathogenesis of osteoarthritis and provides a theoretical basis for the exploration of biomarkers for the diagnosis andtargeted therapy of osteoarthritis.Conclusion 1?The differential expression profiles of LncRNAs and mRNAs in osteoarthritis cartilage cells were constructed by gene sequencing?2?The biological functions of differentially expressed LncRNAs and m RNAs were commented by GO and Pathway analysis.3 ? we validated differentially expressed LncRNAs and m RNAs(3 up and 3 down)by qRT-PCR in more contrasted samples.
