Objective:1.To analyze the rate of positive expression and expression level of autophagy protein LC3-?,KLK6/8 and Claudin-3/4 in the tissues of different serous ovarian tumors.2.To explore the association of the expression level of autophagy protein LC3-?,KLK6/8 and Claudin-3/4 with the FIGO stage,pathological grading and lymphatic metastasis in patients with ovarian serous cystadenocarcinoma.3.To explore the effect of expression level of LC3-?,KLK6/8 and Claudin-3/4 to clinical long-term prognosis in patients with ovarian serous cystadenocarcinoma.Methods:1.A total of 108 patients who were diagnosed pathologically with ovarian serous tumor were selected from January 2015 to January 2017.Among them,there were 42 patients with serous cystadenoma(group A),27 patients with borderline serous tumor(group B),39 patients with serous cystadenocarcinoma.At the same time,we selected 80 patients who were also diagnosed pathologically with ovarian serous cystadenocarcinoma from January 2010 to December 2014.Therefore,group C were constituted with 119 patients with ovarian serous cystadenocarcinoma from January 2010 to January 2017.Control group were 46 patients with normal ovarian epithelial tissue.We tested the rate of positive expression and expression level of autophagy protein LC3-?,KLK6/8 and Claudin-3/4 in the tissues of different ovarian serous tumors and normal epithelial tissue by chemical immunoassay.We used Western blot to detect the content of autophagy protein LC3-?,KLK6/8 and Claudin-3/4 in each group.We analyzed the differences of the rate of positive expression and expression level of autophagy protein LC3-?,KLK6/8 and Claudin-3/4 in the tissues of different ovarian serous tumors and normal epithelial tissue.2.In 119 patients with ovarian serous cystadenocarcinoma,we analyzed the association of expression level of LC3-?,KLK6/8 and Claudin-3/4 with age,tumor size,pathological grading,FIGO stage,menopausal status and lymphatic metastasis.3.All patients with ovarian serous cystadenocarcinoma were followed up for 1 to 8 years.We analyzed the effect of expression level of LC3-?,KLK6/8 and Claudin-3/4 to clinical long-term prognosis in patients with ovarian serous cystadenocarcinoma.4.We used SPSS18.0 software for statistical analysis.Continuous variables were expressed as mean ± standard deviation.Single-factor ANOVA was used to compare in multiple groups,and further comparison between two groups were performed with LSD test.Categorical variables were expressed as counts and percentages(%),and comparison was performed using a chi-square test.Comparison between two groups was performed by chi-square segmentation of line by column table.Two-way ordinal categorical data was performed by association test.Multivariate Logistic regression was used to analyze the risk factors of clinical death.Log-rank test was used to compare the differences of Kaplan-meier survival curves in each groups.Multivariate Cox regression was used to analyze the independent risk factors of long-term prognosis in patients with ovarian serous cystadenocarcinoma.Results:1.The rate of positive expression of autophagy protein LC3-? in control group,group A,group B and group C were 0%,52.4%,92.6% and 95% respectively,the rate of positive expression and expression level of autophagy protein LC3-? in borderline serous tumor group and serous cystadenocarcinoma group were higher than serous cystadenoma group and control group,the differences were statistically significant(P<0.01).The expression level of LC3-? had an increasing tendency along with the increase of grade malignancy of serous tumor(P<0.001).The result of Western blot showed the expression level of autophagy protein LC3-? in different groups was control group < group A < group B < group C,the differences were statistically significant(P<0.05).In patients with ovarian serous cystadenocarcinoma,the expression level of autophagy protein LC3-? was associated with FIGO stage,pathological grading and lymphatic metastasis(P<0.05),and not associated with age of patient,tumor size and menopausal status(P>0.05).2.The rate of positive expression of KLK6 in control group,group A,group B and group C were 0%,23.8%,77.8% and 79.9% respectively.The rate of positive expression and expression level of KLK6 in borderline serous tumor group and serous cystadenocarcinoma group were higher than serous cystadenoma group and control group,the differences were statistically significant(P<0.01).The rate of positive expression of KLK8 in control group,group A,group B and group C were 0%,26.2%,25.9% and 60.5% respectively.The rate of positive expression and expression level of KLK8 in serous cystadenocarcinoma group were higher than borderline serous tumor group,serous cystadenoma group and control group,the differences were statistically significant(P<0.01).The expression level of KLK6 and KLK8 had an increasing tendency along with the increase of grade malignancy of serous tumor(P<0.001).The result of Western blot showed the expression level of KLK6 and KLK8 in different groups was control group < group A < group B < group C,the differences were statistically significant(P<0.05).In patients with ovarian serous cystadenocarcinoma,the expression level of KLK6 and KLK8 was associated with FIGO stage,pathological grading and lymphatic metastasis(P<0.05),and not associated with age of patient,tumor size and menopausal status(P>0.05).3.The rate of positive expression of Claudin-3 in control group,group A,group B and group C were 0.0%,35.7%,51.9% and 79.0% respectively.The rate of positive expression of Claudin-4 in control group,group A,group B and group C were 0.0%,23.8%,37% and 78.2% respectively,the rate of positive expression and expression level of Claudin-3 and Claudin-4 in borderline serous tumor group and serous cystadenocarcinoma group were higher than serous cystadenoma group and control group,the differences were statistically significant(P<0.01).The expression level of Claudin-3 and Claudin-4 had an increasing tendency along with the increase of grade malignancy of serous tumor(P<0.001).The result of Western blot showed the expression level of Claudin-3 and Claudin-4 in different groups was control group < group A < group B < group C,the differences were statistically significant(P<0.05).In patients with ovarian serous cystadenocarcinoma,the expression level of Claudin-3 and Claudin-4 was associated with FIGO stage,pathological grading and lymphatic metastasis(P<0.05),and not associated with age of patient,tumor size and menopausal status(P>0.05).4.All patients with ovarian serous cystadenocarcinoma were followed up for 1 to 8 years.We used multivariate Logistic regression to analyze the risk factors of clincal death,and the result showed that FIGO stage,pathological grading,lymphatic metastasis and the expression level of autophagy LC3-?,KLK6/8 and Claudin-3/4 were the independent risk factors of clinical death.Kaplan-meier survival curves and Log-rank test showed that the median survival time had statistical differences among different expression groups of autophagy LC3-?,KLK6/8 and Claudin-3/4.Multivariate Cox regression analysis showed that pathological grading,FIGO stage,lymphatic metastasis and high expression of autophagy LC3-? and Claudin-3 were the independent risk factors of clinical long-term prognosis in patients with ovarian serous cystadenocarcinoma.Conclusion:1.The rate of positive expression and expression level of autophagy protein LC3-?,KLK6/8 and Claudin-3/4 were associated with grade malignancy of ovarian serous tumor.2.In patients with ovarian serous cystadenocarcinoma,the expression level of autophagy protein LC3-?,KLK6/8 and Claudin-3/4 were associated with pathological grading,FIGO stage and lymphatic metastasis.3.The high expression of autophagy LC3-? and Claudin-3 were the independent risk factors of clinical long-term prognosis in patients with ovarian serous cystadenocarcinoma.The high expression of autophagy protein LC3-? and Claudin-3 in tissue maybe play an important role for the occurrence and development of ovarian serous cystadenocarcinoma.
