| Objective: 1)To study the blood glucose level and its influencing factors of elderly men in Uyghur nationality in some parts of Xinxiang through cross-sectional study based on population;2)To investigate the correlation between testosterone level and sex hormone binding globulin and blood glucose level;3)The relationship between early detection of sex hormone binding globulin gene polymorphism and type 2 diabetes mellitus.Methods: 1)Adopt the method of cluster sampling in Urumqi region Uyghur men older than 40 samples,determination of testosterone,insulin,fasting blood glucose,blood sugar 2hours after OGTT,height,weight and other biochemical and physical index,comparison between type 2 diabetes mellitus and non-diabetic biochemical index difference.The influence factors of type 2 diabetes mellitus were studied by using the non-conditional Logistic regression equation.2)The research object will be normal glucose metabolism level,to determine level of sex hormone binding globulin(SHBG),grouped according to different levels of sex hormone binding globulin(SHBG)and compare the differences in the levels of blood sugar under different levels of sex hormone binding globulin(SHBG)and further study of sex hormone binding globulin(SHBG)influence factors and its correlation with glycosylated hemoglobin;3)Select eight of sex hormone binding globulin(SHBG)gene locus(rs11078701,rs858521,rs858518,rs3760213,rs1799941,rs6257,rs6259,rs727428)polymorphism analysis,and casecontrol study,biochemical index can be divided into case group and control group to learn more about each point correlation between gene polymorphism and type 2 diabetes.Results: 1.The increase of HOMA-IR in middle-aged and elderly Uyghur men was an independent risk factor for type 2 diabetes.High total testosterone levels were protective factors for type 2 diabetes,independent of HOMA-IR,lipid levels,and blood pressure levels.After considering the total testosterone level,vitamin D levels were not associatedwith type 2 diabetes.2.There are differences in the levels of sex hormones,sex hormones and globulin levels in middle-aged and elderly Uyghur men.Although the age difference between different sex hormone binding globulin levels was statistically significant,aging did not become the independent factor of sex hormone binding globulin.The independent influence factors of sex hormone binding globulin were low density lipoprotein,25 OHD,HOMA-IR,testosterone and fasting insulin.Normal glucose metabolism did not see the linear correlation between glycosylated hemoglobin and sex hormone binding globulin level.3.After considering waist circumference,BMI,blood lipid level and age factor,the combination of sex hormone binding globulin level was the independent influence factor of type 2 diabetes.The risk of type 2 diabetes decreased with the increase of the binding protein level.Different genotypes of rs858521 were statistically different in the control group and the type 2 diabetes group,and the allele G was the risk allele.Different genotypes of rs1799941 were statistically different in the two groups,and no allele was found in the healthy control group.Different genotypes of rs6259 were statistically different in the two groups,and the allele G was the risk allele.SHBG gene frequency of haplotype distribution between case group and control group rs6259 rs727428 haplotype GA,GG,frequency distribution between the two groups statistically significant rs858518-rs3760213-rs1799941-rs6257 haplotype TCGC two groups to compare the OR value of0.697,95% CI(0.867 ~ 1.743),the rs6259-rs727428 haplotype GA(OR = 0.468)comparing the two groups,95% CI(0.320 ~ 0.683],rs6259 rs727428 haplotype GG(OR =1.478)comparing the two groups,95% CI(1.053 ~ 2.074).Conclusion: the results suggest that insulin level and homa-ir are independent factors that affect the binding of the sex hormone binding globulin,and may occur before blood sugar reaches the standard of diabetes.Sex hormone binding globulin levels may be an independent protective factor for type 2 diabetes.And SHBG gene haplotype rs6259-rs727428,rs6259 rs727428 haplotype,rs6259 rs727428 haplotype in diabetic and non-diabetic group there are differences in distribution,for the first time,the uighurs sex hormone binding globulin(SHBG)gene polymorphism and type 2 diabetes association,more research is needed to type 2 diabetes susceptibility loci of sex hormone binding globulin(SHBG)gene regulation mechanism is discussed in this paper. |
