| Objective:By studying the effect of"Adjust Zang Dredge Meridian" Electric Acupuncture on body weight,blood glucose,liver histopathology and liver insulin resistance in db/db mice,in order to explore the intervention effect of"Adjust Zang Dredge Meridian" Electric Acupuncture on db/db mice and its effect on liver insulin resistance,and to explain the possible mechanism of"Adjust Zang Dredge Meridian" Electric Acupuncture in improving liver insulin resistance in db/db mice.Methods:22 db/db mice were randomly divided into electroacupuncture group(n=11)and model group(n=11).In the blank group,9 db/m mice were fed under the same conditions in the same period,and there were 3 groups.The electroacupuncture group was treated with electroacupuncture intervention of"regulating viscera and dredging collaterals",once a day for 30 minutes,6 times as a course of treatment,with a rest of 1 day during the course of treatment for 2 consecutive courses.In the course of the experiment,the model group and the blank group did not receive any special treatment,only as a control observation.Before electroacupuncture treatment,1 week after electroacupuncture treatment and 2 weeks after electroacupuncture intervention,blood samples were collected from the tail tip of mice in each group to measure blood glucose and weigh the body weight of mice in each group.At the end of treatment,the samples were taken and the livers of mice were detected.The histopathological changes of liver tissue were detected by HE staining,the content of hepatic glycogen was detected by anthrone method,and the expression of proteins related to insulin signal pathway,including PI3K,p-Akt and p-GSK-3β,were detected by immunohistochemistry and Western Blot.Results:1.General situation of laboratory animals and related indicators(1)General indicators of experimental animals:Blank control group:mice with shiny skin,good condition,sensitive activity,normal food intake and water intake;model group:mice with dull and dull skin,dull state,sluggish state,and slow response,Food intake and water intake were significantly increased;acupuncture group:the fur of the mice was dull,the state was normal,the response was slightly dull,and the food intake and water intake were increased compared with normal control mice.Water consumption:After 1 week and 2weeks of intervention,the water consumption in the acupuncture group and the model group was significantly increased compared with the blank group during the same period(***P<0.001);a typical important factor for a mouse model of diabetes Symptoms,the water consumption of mice in the model group increased more significantly.Compared with the model group,the increase in water consumption in the acupuncture group was significantly lower than that in the model group(△△△P<0.001).Food intake:After 1 week and 2 weeks of intervention,the food intake of the acupuncture group and the model group increased significantly compared with the blank group during the same period(***P<0.001);this is also a significant feature of the diabetic mouse model,and The food intake of mice in the model group increased more significantly;compared with the model group in the same period,the food intake of the acupuncture group was significantly lower than that in the model group(△△△P<0.001).2.The result of blood glucose changes in each group of mice:Before intervention,there was a significant difference in blood glucose between the blank group,acupuncture group and model group(**P<0.01),and there was no difference in blood glucose between the acupuncture group and the model group(P>0.05);After 1 week of intervention,there was no difference in blood glucose between the acupuncture group and the model group(P>0.05),but it can be seen that the blood glucose of the acupuncture group increased slowly compared to the model group;after 2 weeks of intervention,the Compared with the group,the blood glucose change of the acupuncture group had a significant improvement(△P<0.05).Test results of liver glycogen content of mice in each group:Compared with the mice in the blank group,the liver glycogen content of the mice in the acupuncture group was reduced,but there was no significant difference in liver glycogen changes between the two groups(P>0.05).Model group Compared with the mice in the blank group,the liver glycogen content was significantly reduced,and the changes were significantly different(P<0.05).3.The results of liver morphological changes of mice in each group:in the blank group,the liver tissue structure was normal,the cells arranged regularly,the cytoplasm was uniform,the structure of hepatic lobules was normal,the hepatocytes distributed in strips around,and there was no inflammatory cell infiltration.In the model group,the arrangement of hepatocyte cord was disordered,most hepatocytes showed severe steatosis,hepatocyte swelling,lipid droplet vacuoles of different size and number could be seen in the cytoplasm,and some cells showed edema.The arrangement of hepatocytes in the electroacupuncture group was more uniform and orderly,and the structure was more complete.The phenomena of cell edema,steatosis and vacuolation in the electroacupuncture group were significantly better than those in the model group,but the overall condition was not as good as that in the blank group.Results of PI3K,p-Akt,and p-GSK-3βprotein expression in liver tissue of mice in each group:Compared with the blank group,the protein expression levels of PI3K,p-Akt,and p-GSK-3βin liver tissue of the model group were significant.Decreased(△P<0.05);Compared with the model group,the protein expression levels of PI3K,p-Akt,and p-GSK-3βin the liver tissue of the"Adjust Zang Dredge Meridian" Electric Acupuncture group were significantly increased(#P<0.05).Conclusion:1."Adjust Zang Dredge Meridian" Electric Acupuncture can improve the general symptoms and body weight of db/db mice,reduce the level of blood glucose,and has a therapeutic effect on type 2 diabetes.2."Adjust Zang Dredge Meridian" Electric Acupuncture can improve the level of glucose metabolism in db/db mice.3."Adjust Zang Dredge Meridian" Electric Acupuncture can improve and repair the liver tissue of db/db mice.4."Adjust Zang Dredge Meridian" Electric Acupuncture may improve the level of insulin resistance in the liver by increasing the expression of PI3K,p-Akt and p-GSK-3βin the liver,and play a therapeutic role in the treatment of type 2 diabetes. |
