| Objective: Study on Jiedu Tongluo Yishen Concentrated Pills on glucose metabolism,lipid metabolism,renal function,renal histopathology,glucagon-like peptide 1 serum in experimental type 2 diabetes rats Effects of concentration,GLP-1 receptor gene expression,and cAMP / PKA pathway-related protein expression,preliminary exploration of its protective mechanism of the kidney,and verifying the scientificity of the theory of "toxic damage to the kidney network" by Professor Nan Zheng Provide new ideas for the treatment of diabetic nephropathy by Chinese medicine.Methods:1.60 male SPF-grade SD rats were adaptively fed for 1 week.After randomization,high-fat diet combined with STZ intraperitoneal injection was used to induce a diabetic(DM)rat model.Blood was collected after 72 hours,and fasting blood glucose was ≥16.7mmol / L is regarded as a successful model.The 45 successful models were randomly divided into 9 DM model groups,high-dose,medium-dose,and low-dose groups of Jiedu Tongluo Yishen Concentrated Pills and 9 in the positive control group(irbesartan tablet).The traditional Chinese medicine high,middle and low dose groups were administrated with 3.0g/kg/d,1.5g/kg/d,0.75g/kg/d Jiedu Tongluo Yishen Concentrated Pill Suspension;the positive control group was given Irbe Satan tablet suspension 13.5mg / kg / d was administered to the stomach,and it was observed continuously for 12 weeks.Observe the general state of the rats,record the body weight and urine output for 24 hours;record the kidney weight / body weight at the end of the experiment;measure fasting blood glucose,glycated hemoglobin,and 24 h urine microalbumin;measure renal function and blood lipids.2.Observe the pathological and morphological changes of renal tissue by using light microscope and transmission electron microscope to study the effect of Jiedu Tongluo Yishen Concentrated Pill on the pathological and morphological characteristics of kidney tissue in diabetic rats.3.ELISA method,immunohistochemical method,Westenblot,RT-PCR method can beused to detect Jiedu Tongluo Yishen Concentrated Pills in renal tissue serum GLP-1concentration,renal tissue GLP-1R related protein can also be similar.4.ELISA,immunohistochemistry and Westenblot were used to observe the effects of cAMP,8-OHdG,PKA and P47-phox.Results:1.Jiedu Tongluo Yishen Concentrated Pills can alleviate the common clinical symptoms of experimental DM rats such as malaise,polydipsia,polydipsia,polydipsia,weight loss and so on.2.Jiedu Tongluo Yishen Concentrated Pill can improve glucose metabolism and lipid metabolism,and reduce fasting blood glucose,TC and TG concentrations in experimental DM rats.3.Jiedu Tongluo Yishen Concentrated Pill can inhibit early proteinuria in experimental DM rats,reduce Scr and BUN,and protect renal function.4.Jiedu Tongluo Yishen Concentrated Pill has the effect of improving the pathological damage of kidney tissue structure in experimental DM rats,and has the effect of delaying the pathological changes of kidney tissue.5.Jiedu Tongluo Yishen Concentrated Pill can increase the serum GLP-1 concentration in experimental DM rats and up-regulate the effects of renal GLP-1R receptor protein and gene expression.6.Jiedu Tongluo Yishen Concentrated Pills can regulate the content of cAMP,PKA,8-OHdG,p47-phox in the kidney tissue of experimental DM rats,activate the cAMP / PKA pathway,down-regulate 8-OHdG in urine,and play an antioxidant role.The role of stress.Conclusion: Jiedu Tongluo Yishen Concentrated Pill can improve the general state of experimental DM rats,improve the metabolism of glucose and lipids,and inhibit the pathological damage of renal tissue structure.It has the effect of increasing serum GLP-1concentration and GLP-1R expression in renal tissue and activating The cAMP / PKA pathway down-regulates 8-OHdG and plays an antioxidant role.Professor Nan Zheng,the mentor,proposed that the key to the pathogenesis of thirst-kidney nephropathy is "toxicdamage to the kidney collaterals,evil membrane membrane".Under the guidance of this theory,this experimental study combined the treatment of "Jiedu Tongluo Yishen Guixie" and "Tiao San Gao,Da Mo Yuan" with the modern medical pathogenesis of diabetic nephropathy,and found that Jiedu Tongluo Yishen Concentrated pills may directly and indirectly protect the kidneys by regulating GLP-1. |
