Study On Function And Mechanism Of Long Noncoding RNA LINC00942 In CCRCC | | Posted on:2021-02-15 | Degree:Doctor | Type:Dissertation | | Country:China | Candidate:X S Wang | Full Text:PDF | | GTID:1364330611491574 | Subject:Surgery | | Abstract/Summary: | PDF Full Text Request | | Background: Renal clear cell carcinoma(RCC)has been widely concerned and studied as a common tumor of Urinary system swelling.According to the WHO classification and investigation of RCC,RCC is the most common type of RCC,with a low 5-year survival rate and the worst prognosis in RCC.With the development of second-generation sequencing technology and high-throughput gene detection technology,the mechanism of gene regulation of tumor occurrence and development is getting more and more attention.The purpose of this study is to use bioinformatics data to predict the genes that affect the prognosis and cell characteristics of renal clear cell carcinoma,and to confirm its function and mechanism by experiments,so as to provide basis and reference for clinical diagnosis and treatment of renal clear cell carcinomaMethods: The clinical data and expression data of patients with renal clear cell carcinoma were obtained by online database.GSEA was used to analyze the gene set that may play a role and screen the genes.Using the methods of batch survival analysis,multiple factor regression analysis and differential expression analysis,the genes that may affect the survival and prognosis of patients were screened out.Through the online website,we can predict the possible upstream regulatory genes and further predict the possible target genes of the upstream regulatory genes,and analyze the possible roles and ways of action through the experimental verification.QRT-PCR was used to detect the expression of long noncoding RNA LINC00942 and mi R-3139 and the possible way of their mutual regulation.The regulatory effect of long noncoding RNA LINC00942 and mi R-3139 on renal clear cell carcinoma cells was verified by cytological study.Western blotting was used to verify the changes of genes in the middle and lower reaches of renal clear cell carcinoma cells.Dual Luciferase Report experiment verified the regulatory model between long noncoding RNA and its downstream target genes.Finally,the effect of LINC00942 was verified in vivo by animal experiments.Results: 1.The risk assessment model constructed by m RNA IL-2,NCR-2,MYH7 and KIR3DL2 can effectively analyze and predict the prognosis of patients with renal clear cell carcinoma.2.Long noncoding RNA LINC00942 has high expression in renal clear cell carcinoma cell line and renal clear cell carcinoma tissue.3.LINC00942 can regulate the proliferation,migration,invasion and metastasis of renal clear cell carcinoma cells.4.LINC00942 can participate in the regulation of m RNAs in the middle and lower reaches of renal clear cell carcinoma cells,and then regulate the cytological characteristics of renal cancer cells.5.Micro RNA mi R-3139 can regulate the expression of LINC00942,and it can regulate the downstream m RNA GPM6 A and HIF mediated signal pathway.6.LINC00942 interacts directly with mi R-3139.7.Animal experiments have proved that LINC00942 can affect the tumorigenicity of renal clear cell carcinoma cells and the growth rate of tumors in vivo.Conclusion: 1.The over expression of IL-2,NCR-2,MYH7 and KIR3DL2 is significantly related to the survival and prognosis of patients with renal clear cell carcinoma.It can be used as an independent risk factor for evaluating survival and prognosis of patients with renal clear cell carcinoma.2.The new risk assessment model can be used to predict the survival and prognosis of patients with renal clear cell carcinoma.3.Our findings may provide a new clinical strategy for the early diagnosis and treatment of renal clear cell carcinoma.4.The long noncoding RNA LINC00942 may regulate and influence the occurrence of renal clear cell carcinoma in the way of endogenous competitive inhibition,and then affect the survival and prognosis of patients with renal clear cell carcinoma.5.LINC00942 is highly expressed in renal clear cell carcinoma cells and tissues.6.LINC00942 can regulate the proliferation,migration,invasion and metastasis of renal clear cell carcinoma.7.LINC00942 can effect the cytological characteristics of renal clear cell carcinoma in the way of mi R-3139.8.LINC00942 can provide new tumor markers and targets for clinical diagnosis and treatment of renal clear cell carcinoma. | | Keywords/Search Tags: | Renal clear cell carcinoma, GSEA, proliferation, migration, invasion and metastasis, Long noncoding RNA | PDF Full Text Request | Related items |
