Objectives:Thyroid cancer is the most common malignant endocrine tumor.The tissue types of thyroid cancer include follicular and par follicular cell types(also known as C cell types).Follicular thyroid cancer can be classified into differentiated(DTC)?undifferentiated(UTC)and anaplastic(ATC)thyroid cancers.DTC include papillary(PTC),follicular(FTC),hurtle cell thyroid cancer,and poorly differentiated thyroid cancer(PDTC).With the continuous development of society,the incidence of thyroid cancer patients in China and the world is increasing,and thyroid cancer has become a serious threat to human life and health.Research on genes that regulate the malignant biological behavior of thyroid cancer will help to reveal the pathogenesis of thyroid cancer.LncRNA is a class of RNA transcripts with a length greater than 200 bp,which has higher temporal and spatial specificity and lower interspecies conservation than m RNA.Most lnc RNAs are located in the nucleus,but some lnc RNAs also play a role in the cytoplasm.LncRNA was previously thought to be a "garbage" product with no biological function and protein coding ability.However,more and more studies are currently showing that lnc RNAs are involved in the pathogenesis of many diseases.Studies have shown that lnc RNA plays an important role in tumorigenesis,angiogenesis,proliferation,migration,apoptosis and differentiation of malignant tumors.To date,a large number of tumor-associated lnc RNAs have been identified and demonstrated to play a key role in tumorigenesis.Therefore,it is of great significance to study the mechanism of lnc RNA affecting tumorigenesis and development.The lnc RNA H19 is located at the p15 site of chromosome 11,and is a gene expressed by the mother line,which plays a vital role in the development of mammals.Studies have shown that lnc RNA H19 is associated with a variety of malignancies,such as lung cancer,gastric cancer,colon cancer,pancreatic cancer,ovarian cancer,neuroblastoma,bladder cancer,breast cancer,and esophageal cancer.There are few studies on the relationship between lnc RNA H19 and thyroid cancer.LncRNA H19 can inhibit the proliferation of a variety of malignant tumor cells,which plays an important role in the process of tumor cell apoptosis.This study examined the expression level of lnc RNA H19 in differentiated thyroid cancer tissues and analyze the relationship between lnc RNA H19 and clinicopathological parameters of patients with differentiated thyroid cancer.MTT assay and flow cytometry were used to observe the effect of lnc RNA H19 on proliferation and apoptosis of differentiated thyroid cancer cells.RT-q PCR and Western blot were used to observe the relationship between lnc RNA H19 and apoptosis-related proteins and PI3K/AKT signaling pathway.To further elucidate the mechanism of action of lnc RNA H19 on the malignant biological characteristics of differentiated thyroid cancer,and to provide experimental evidence for finding new targets for the treatment of differentiated thyroid cancer.Methods:1.Identification of lnc RNA H19 expression level in differentiated thyroid cancer The m RNA expression levels of lnc RNA H19 were detected by quantitative real time PCR in differentiated thyroid cancer tissues2.Effect of lnc RNA H19 on proliferation and apoptosis of differentiated thyroid cancer cells Transfection of lnc RNA H19 si RNA knocks down lnc RNA H19 in differentiated thyroid cancer cell lines FTC-133 and TPC-1,and the cell transfection efficiency was detected by real-time PCR.The effect of knockdown of lnc RNA H19 on the proliferation of differentiated thyroid cancer cells was detected by MTT assay.The effect of knockdown of lnc RNA H19 on apoptosis of differentiated thyroid cancer cells was observed by flow cytometry.3.The molecular mechanism of lnc RNA H19 on apoptosis of differentiated thyroid cancer cells Real-time PCR was used to detect the effect of knockdown of lnc RNA H19 on the expression of apoptosis-related molecules Bax,Bcl-2 and caspase-3 in differentiated thyroid carcinoma FTC-133 and TPC-1 cells.The effect of knockdown of lnc RNA H19 on the expression of apoptosis-related proteins Bax,Bcl-2 and caspase-3 in differentiated thyroid carcinoma FTC-133 and TPC-1 cells was observed by Western blot.The effect of knockdown of lnc RNA H19 on PI3 K / AKT signaling pathway was observed by Western blot.Results:1.The expression of lnc RNA H19 was significantly upregulated in differentiated thyroid cancer tissues.2.Knockdown of lnc RNA H19 inhibited the proliferation of differentiated thyroid cancer cells and promoted the apoptosis of differentiated thyroid cancer cells.3.Knockdown of lnc RNA H19 increased the expression of Bax and Caspase-3 m RNA and protein in differentiated thyroid carcinoma FTC-133 and TPC-1 cells,and inhibited the expression of Bcl-2 m RNA and protein.4.Knockdown of lnc RNA H19 in differentiated thyroid cancer cells significantly inhibited the expression of p-PI3 K and p-AKT,indicating that lnc RNA H19 can affect the apoptosis of differentiated thyroid cancer cells through PI3 K / AKT signaling pathway.Conclusions:The expression of lnc RNA H19 is significantly increased in differentiated thyroid cancer tissue,which can affect the cell proliferation and apoptosis level of differentiated thyroid cancer cells by affecting the PI3 K / AKT signaling pathway.
