BackgroundDepression is a kind of complex emotional mental disorder,which includes many kinds of mental and physical symptoms.The main clinical symptoms are marked and lasting low mood,decreased activity ability and slow thinking and cognitive function.Because of the high prevalence,disability and recurrence rate,the National Mental Health Work Plan(2015-2020)has been listed as a key disease by the National Health Commission.In the category of TCM,"depression" is emotional disease,its symptoms and depression syndrome,lily disease,dirty and so on have similarities,and emotional depression of "depression" is more consistent.For the clinical treatment of depression,generally drugs-based,psychological and other therapies as a supplement,for the symptoms of mild patients,cognitive and behavioral therapy can be applied.Traditional Chinese medicine has many advantages in treating depression,such as traditional Chinese medicine,acupuncture,emotional intervention and so on.As a psychological intervention method with Chinese medical characteristics,the psychological therapy of emotional competition is to activate and mobilize the specific emotional experience of patients consciously in order to restrict and eliminate the corresponding morbid emotions,to improve the symptoms of depression,improve the efficacy of a positive role."Delighting Adjusting Sorrow" is a kind of guiding ideology and specific measures of intervening depression from the idea of traditional Chinese medicine.In short,"Delighting Adjusting Sorrow" is a positive,positive emotional constraints,regulation of negative,negative emotional process.According to TCM theory,"Delight" can alleviate the intensity and speed of Qi movement,so as to avoid the vicious circle of Qi depression caused by melancholy,Qi Depression and melancholy,and alleviate the symptoms of "Qi Depression" in the early stage of mild depression or depression.However,the research on the mechanism of its action is mostly based on the theory of Qi mechanism change and Wu Xing,and the exploration on the level of cognitive neural mechanism is rare.Objective1 Develop a standardized video database of "happy" and "sad" emotions.2 Prove the hypothesis that the activation of "Delighting" could adjust the negative cognitive processing bias of patients with depression3 Explore the characteristics of ERP components(P2,N2)of depression patients in the emotional face forcing task after "Delighting" activation.Methods1.Study one developed a standardized "happy" and "sad" emotional activation video material library.The material library includes 21 videos that can induce "happiness",8 videos that can induce "sadness" and 13 neutral videos that can induce calm emotion.The results show that the video library has high reliability and validity,the selected video can induce target emotion more accurately,and the intensity of emotion is more reasonable,each video contains target emotion type,duration,hit rate,emotional intensity potency,arousal,motivational intensity and other parameters,and the scores of three dimensions of Valence,arousal,and motivational intensity are different for different types of video,with some degree of differentiation.To sum up,the video library can provide experimental stimulus materials of "happiness","sadness" and neutral emotion activation,including more accurate parameters of emotion activation dimension,which can provide a standardized tool for TCM emotion-related experimental research.2.In Study 2,the standardized video database of "happy" and "Sad" emotions developed in study 1 was used as a tool to explore the effect of "happy" activation,the differences between depression patients and healthy controls in ERP components(P2,N2)and behavioral outcomes were compared in order to explain the mechanism of "Delighting Adjusting Sorrow" regulating negative cognitive processing bias in depression patients.In the second study,the depression group was recruited by posting posters and the introduction of clinical doctors,while the health control group was recruited by posting posters,Wechat and other online channels,and according to the Depression Group participants match gender,age and other demographic information.The ERP experiment design is:Random Group of depression patients and healthy subjects to "happy" neutral emotional activation,subjects were divided into 4 groups:Depression "Delighting" Activated Group,Depression Not Activated Group,Health Control"Delighting" Activated Group,Health Control Not activated group,using ERP technology,to observe the difference of emotional forcing response to mosaic neutral faces,the behavioral judgment of Target Stimuli(mosaic neutral faces)and the ERP components(P2,N2,etc.)were used as the outcome indexes,to clarify the role of "Delighting Adjusting Sorrow" in controlling negative emotion and enhancing negative emotion in emotion-related experimental research.2.The study included 89 subjects,including 44 depressive patients,20 male,24 female,45 healthy control subjects,22 male,23 female.The results of emotion forced selection showed that the judgment times of positive and negative emotion expression of target stimulus were significantly different among different groups,and the main effect was significant among depression health control groups,the main effect of emotion activation was also significant,and there was no interactive effect between depression health group and different emotion activation.For patients with depression,the number of negative emotion judgment after neutral emotion activation was significantly higher than that of positive emotion judgment,but there was no significant difference between positive and negative emotion judgment when happy emotion was activated.For the healthy control group,there was no significant difference in the judgment of positive and negative emotion under neutral emotion condition,the number of times that the target stimulus was judged as negative emotion was significantly less than that of positive emotion.The results of reaction time showed that the main effect of emotion activation mode and depression health group were not significant,and there was no interactive effect between depression health group and different emotion activation.The statistical results of P2 latency showed that there were some differences in different brain regions in each group.The main effect of depression and Health Group in the four brain regions was significant,in the left frontal region,right frontal region and top region,the depression group was larger than the healthy control group,in the Occipital region,the depression group was smaller than the Healthy Control Group;The main effect of emotion activation was not significant in the four brain regions,and the interaction between depression health group and different emotion activation was not significant in the four brain regions.In the left frontal,right frontal and top regions,the latency of P2 in each group was higher than that in the healthy control group In patients with occipital depression,the activation of liking and neutral emotion were lower than that of healthy control group.The statistical results of P2 wave amplitude showed that there were some differences in different brain regions in each group.The main effects of depression and health group in the four brain regions were significantly less than that of healthy control group,and the main effects of emotion activation in the right frontal region and the top region were significantly less than that of neutral emotion activation The interaction between depression health group and different emotion activation was not significant in the four brain regions.The results of the comparison of the amplitude of P2 components in different brain regions in each group were as follows:In the left frontal region,right frontal region and the top region,the positive and neutral activation were less in the depressed patients than in the healthy ones,patients with depression also had less neutral activation than healthy subjects,and patients with depression had less neutral activation than healthy subjects in occipital region.The statistical results of N2 latency showed that there were some differences in different brain regions in each group.The main effects of depression and health were significant in occipital region,but not in left frontal region,right frontal region and top region,and the main effects of emotional activation were not significant in four brain regions The interaction between depression health group and different emotion activation was not significant in the four brain regions.N2 component latency in occipital region,the activation of liking and neutral emotion in depressive patients were lower than those in healthy control group.The statistical results of N2 wave amplitude showed that there were some differences among different brain regions in each group.The main effect of depression and Health Group in the four brain areas was significantly smaller than that in the healthy control group,and the main effect of emotional activation was not significant in the four brain areas The interaction between depression health group and different emotion activation was not significant in the four brain regions.Compared with the amplitude of N2 components in different brain regions of each group,the activation of liking and neutral emotion in depressive patients were lower than that in healthy group.Conclusion1.In this study,a standardized emotional activation video database was developed,which included experimental stimuli of "happiness","sadness" and neutral emotional activation.Each video had more accurate emotional activation parameters,it can provide a standardized tool for the experimental research of TCM emotion.The database includes 21 "happy" video clips,8 "Sad" video clips and 13 neutral video clips.The results show that the video library has high reliability and validity,and it can induce target emotion accurately.Each video contains target emotion type,duration,hit rate,emotional intensity potency,arousal,motivational intensity and other parameters,and the scores of three dimensions of Valence,arousal,and motivational intensity are different for different types of video,with some degree of differentiation.2.The study confirmed the existence of negative cognitive bias in patients with depression,which was supported by the results of emotional forced choice,P2 and N2 components.Patients with depression were more likely to judge the emotional attributes of neutral stimuli as negative than those of healthy subjects.The P2 component results showed that depressed subjects were more sensitive to visual activation of face images than healthy subjects,and that P2 peaks appeared earlier,but at higher levels of thought judgment,depressed subjects responded later than healthy subjects,and when paying attention to facial expression,less attention resources were allocated actively.N2 showed that depression patients were more sensitive to the inhibition of visual activation of face images and less able to control the emotional response to face images.The negative cognitive bias of depressive patients has appeared in the early stage of passive visual stimulus processing,and also in the middle stage of attention process and cognitive resource allocation.3.The behavioral statistics indicate that,the degree of negative cognitive bias was reduced in patients with depression after the activation of "Delighting",while healthy people were more inclined to make positive emotional judgments about uncertain neutral stimuli.In ERP data,the results of P2 component showed that P2 decreased slightly after the activation of "Delighting" in depressive patients,which was marginal and significant,possibly related to the decrease of attention resource allocation to facial expression.There was no significant difference in N2 latency and amplitude between happy activation and neutral emotional activation.
