Objective:1.To study the expression level of NDRG1 gene in different types of childhood acute leukemia(AL)and explore its significance.2.To investigate the expression and relationship of NDRG1 gene,transferrin gene and ferritin gene in children with acute myeloid leukemia(AML)after formal treatment.Methods:1.60 patients with acute leukemia(AL)diagnosed from February 2015 to May 2017 were selected and their bone marrow samples were collected,26 cases of acute lymphoblastic leukemia(ALL)and 34 cases of acute myeloid leukemia(AML),including: M1 type 6 cases,M2 type 7 cases,M3 type 5 cases,M4 Type 8 and M5 8 cases.According to the criteria of sample collection and early AL treatment,it was divided into initial group,remission group and non-remission group.43 cases in the initial group,14 cases in the remission group and 3 cases in the non-remission group.AML were treated with DAE chemotherapy regimen,in which M3 was all-trans retinoic acid(ATRA)and soft red.DNR combination therapy.ALL was treated with VDLD chemotherapy regimens.In addition,20 children with idiopathic thrombocytopenia(ITP)were selected as control group.Reverse Transcription Real-Time Polymerase Chain Reaction(RT-PCR)was used to detect the expression of NDRG1 m RNA in the above 80 children.According to the chronological order of selection,the first 5 children with ALL and AML who were first selected were used.The expression of NDRG2,NDRG3 and NDRG4 m RNA was detected by RT-PCR.2.10 children with AML who were treated were selected and their bone marrow samples were collected,including 8 patients in the remission group and 2 patients in the non-remission group.The expression levels of NDRG1 m RNA,Tf R m RNA,and FN m RNA were detected by RT-PCR.To investigate the relationship between NDRG1 gene expression level and Tf R and FN gene expression levels in children with AML.Results:1.The positive expression rate of NDRG1 in 60 children with AL was 66.7%(40/60).Among them,the positive expression rate of AML was the highest(88.2%),which was significantly higher than that of ALL(38.5%)and control group(10%)(P<0.05),the difference between the latter two was not statistically significant(P> 0.05).2.The relative expression levels of NDRG1 genes of different types of AML were ranked from large to small: M5>M4>M1 and/or M2,and no expression in M3 type was found.3.The NDRG1 gene has a moderately high positive expression in high-risk children with AML.The expression of NDRG1 gene in the initial group of AML was significantly higher than that in the remission group and the control group(P<0.05).The expression of NDRG1 gene was significantly higher in the non-remission group than in the remission group and the control group(P<0.05).The remission group and the control group There was no statistical difference between the groups(P>0.05).4.The positive expression rates of NDRG1 in high-risk group,middle-risk group and low-risk group were 100%,91.7% and 84.6%,respectively.The relative expression levels of NDRG1 gene were 0.61±0.01,0.46±0.08 and 0.31±0.06,respectively.The relative expression of NDRG1 gene was higher in the low-risk group and the middle-risk group(P<0.05).5.The expression of NDRG1?4 gene in bone marrow of children with AL was different.The expression of NDRG1 and NDRG2 in acute leukemia was statistically significant(P<0.05).The expression of NDRG1 gene in AML group was significantly higher than that in ALL group(P<0.05).6.The expression levels of NDRG1 gene in ITP group,remission group and non-remission group were 0.08±0.03,0.17±0.06 and 0.64±0.03,respectively.The expression level of the non-remission group was higher than that of the remission group,and the expression level of the remission group was higher than that of the ITP group(all P<0.001).The expression levels of Tf R gene in ITP group,remission group and non-remission group were 0.37±0.05,1.84±0.11 and 1.41±0.02,respectively.The expression levels of FN gene in ITP group,remission group and non-remission group were 0.25±0.01,0.39±0.05 and 0.61±0.12,respectively.The results showed that the expression level of Tf R gene remission group was higher than that of non-remission group,and the expression level of non-remission group was higher than that of ITP group,the difference was statistically significant(P<0.001).The expression of FN gene in the non-remission group was higher than that in the remission group,and the expression in the remission group was higher than that in the ITP group,and the difference was statistically significant(P<0.001).Conclusion:1.Overexpression of NDRG1 gene is closely related to the incidence of acute leukemia in children.The expression of NDRG1 gene in AML is significantly higher than ALL.2.High expression of the NDRG1 gene in children with AML may indicate a poor prognosis.3.NDRG1,Tf R and FN genes were expressed in AML bone marrow cells.4.There is a certain correlation between the expression level of NDRG1 gene and the balance of iron storage in AML.5.In acute myeloid leukemia,the up-regulation of NDRG1 gene expression may be a direction in the treatment of iron chelators to improve the condition.
