As an initial step toward obtaining a more complete view of the local internal motions of nucleic acids and the role these motions play in protein recognition, we have used solution and solid state NMR conjointly to probe the internal localized motions of a DNA dodecamer that contains a recognition site for the HhaI methyltransferase. Our results strongly suggest that mus-ns motions contribute to the functionally relevant dynamic properties of nucleic acids during DNA methylation, and that the methylation itself strongly affects the dynamics which contribute to function and recognition. |
