Abstract/Summary:
Tissue Engineering aims to restore tissue functions by developing engineered scaffolds providing optimum environment for tissue regeneration. In this dissertation, hybrid tissue scaffolds are modeled, designed and fabricated to control release kinetics spatially and temporarily for improved tissue regeneration. Heterogeneous porous tissue scaffolds with varying characteristics to mediate the release of base material and enclosed biomolecules are proposed based on tissue engineering requirements. Firstly, a novel computer-aided biodegradation model is proposed to control biodegradation by varying the micro-pattern architecture of polymeric tissue scaffolds. Next, scaffolds are biomimetically designed with controlled porosity and optimized internal architecture to enhance fluid transport with continues base material deposition. Then, imaging techniques are applied to explore natural distribution of microspheres encapsulated in tissue scaffolds. The obtained distribution characteristics are mimicked by the developed stochastic modeling study. The study is extended for the development of 3D hybrid wound scaffolds for spatial control of microspheres and base material to synchronize the release kinetics with tissue regeneration. A pressure-assisted multi-chamber single nozzle solid freeform fabrication (SFF) technique is used to fabricate hybrid scaffolds.
