Wingceltis(Pteroceltis tatarinowii)bark of 3-5 years old is an important raw material of Xuan paper,and the bark characteristics directly affect the quality of the paper.It is worthful to investigate the relationship between the bark characteristics and environmental factors,as well as the molecular regulation mechanism of bark characteristics formation.We carried out the bark characteristic analysis,correlation analysis between the bark characteristics and environmental factors,transcriptome analysis and real-time RT-PCR.The following results were obtained.1.The relationship between the bark characteristics of Wingceltis from 11 populations in 7 provinces and ecological environment factors in the collecting locations was determined by redundancy analysis(RDA).The results showed that the bark quality is influenced by environmental factors,and the bark cellulose content,lignin content,fiber length and width in different populations have significant difference;the cellulose,pentosan lignin contents and fiber width are influenced by climatic and soil factors,the higher annual average temperature and the longer the frost-free period are,the higher the cellulose,lignin and pentosan content are and the wider the fiber width is.Moreover soil organic matter and alkali nitrogen content are higher,the four indexes are greater;while the fiber length is mainly with the soil available K factor,climate factors hardly work.2.The results were obtained through transcriptome analysis of Wingceltis phloem from three populations(XA,JX,AL).Wingceltis phloem transcriptome of three populations had certain gene expression differences,the transcriptome expression mode of AL and JL populations is more close in comparison with XA expression mode.The certain expression differences existed in genes of the three population which encode enzymes involved lignin and cellulose biosynthesis.There had various factors influencing Wingceltis lignin and cellulose synthesis,such as the regulation of plant hormones and transcription factors,which showed a complex metabolic regulatory network diagram.3.The followed results was found through real-time RT-PCR analysis of 4 genes encoding enzymes involved cellulose synthesis pathway.4 genes expression in the three populations did not differ significantly during the detection period.Ces A1 and Ces A8 expression existed mainly in the stem,thus we speculate this two genes may involve in secondary cell wall formation.4.The followed results was found through real-time RT-PCR analysis of 3 genes encoding enzymes involved lignin synthesis pathway.PAL and CCoAOMT genes expression in the three populations had no significant difference,but CAD expression in the 3 populations was significantly different.Three genes expression mode of young branches tends to be consistent at different periods,which may be caused by similar regulatory mechanism.PAL expression had both tissue-specific and growth stage specificity,and the expression level was higher in leaves and stems,and reached the peak of expression in May.CCoAOMT gene expressed specially in tissues with high degree wood,and sustained high expression during June,July and August.
