Myzus persicae and aphid transmitted virus disease such as potato Y virus disease(PVY)are important pest and diseases in tobacco,which occur seriously all year round and are difficult to control,causing huge losses to tobacco production.The main purpose of this study is to identify the species of aphids and aphid transmitted virus diseases in tobacco,analyze the genetic diversity of aphid transmitted virus diseases,and study the epidemic correlation between aphids and aphid transmitted virus diseases,and the green control technology of aphids and aphid transmitted virus diseases,so as to provide theoretical basis and application mode for the effective control of the occurrence and damage of aphids and aphid transmitted virus diseases.The main results are as follows:1.Identification of tobacco aphid species and study on its transmitted virus mode.The identification of tobacco aphid species was carried out.It was clear that the main aphid species on tobacco in Anhui Province were M.persicae.The study on the transmitted virus mode of M.persicae showed that the stylet,head and abdomen could carry the virus.The duration of the virus in M.persicae could be up to 10 hours.The transmitted virus efficiency of M.persicae was not directly proportional to it's carrying rate.2.Detection,damage and genetic diversity of aphid transmitted virus to tobacco.In this study,670 tobacco samples were detected by ELISA,PCR and high-throughput sequencing of siRNA,and PVY was identified as the main aphid transmitted virus in Anhui Province.We carried out the detection of plants with virus around the tobacco field and the detection of virus-carrying rate of tobacco seedlings in the greenhouse,and made it clear that the virus-carrying crops mainly include Brassica napus L.,study the epidemic correlation between aphids and aphid transmitted virus diseases,and the green control technology of aphids and aphid transmitted virus diseases,so as to provide theoretical basis and application mode for the effective control of the occurrence and damage of aphids and aphid transmitted virus diseases.3.Screening of new virus transmitted by aphid with high-throughput sequencing of siRNA.A new tobacco virus TV2,was found by high-throughput sequencing of siRNA.The total sequence of TV2 is 5979 nucleotides,which has the highest homology with PLRV(87%).That is the complete genome sequence of a new virus of polerovirus was first reported.The Brassica yellow virus(BrYV)and Chilli veinal mottle virus(ChiVMV)were first detected in Anhui tobacco area.The complete genome sequence of Br YV-AH isolate was analyzed,the total length is 5678bp and encodes six open reading frames.It is a recombinant virus.4.Study on the epidemic correlation between aphids and aphid transmitted virus diseasesA study on the correlation between dynamic migration monitoring of winged aphid and the occurrence of aphid virus disease was carried out.The results showed that the number and time of migration of winged aphid were positively correlated with the occurrence and severity of aphid virus disease.The larger the number of alatae migration,the more serious the aphid transmitted virus disease is;The migration peak of winged aphid was positively correlated with the peak of the virus.After the aphid moved to the tobacco field and transmit the virus,there was a hidden disease process to the virus-carrying tobacco.The duration of this process was mainly related to the climate factors and tobacco growth period.If the weather conditions are favorable for the occurrence of the disease,the cryptic period is short.If the climate conditions were unfavorable for the occurrence of the disease,the hidden disease process could be more than 15-20 days.5.Research and application of green control technology for aphids and aphid transmitted virus diseasesThe research and application of green control technology for pest control and disease prevention were carried out,including natural enemies(Aphidius gifuensis),entomopathogenic fungi and physical methods(non-woven covering,yellow card,etc.)to control M.persicae,new immune inducer to prevent aphid transmitted virus diseases,etc.The following green control technologies with better control effect were selected:the average control effect of A.gifuensis on M.persicae was 75%,the average reduction of aphid control pesticides in production season was three times and virus control pesticides was one time,the average reduction of chemical pesticide input was 6.4 yuan/667m~2,and application cost was 12 yuan/667m~2.Compared with the surrounding conventional control areas,the relative control efficiency of non-woven covering demonstration areas,yellow card aphid induction demonstration areas and immune inducers for virus disease prevention demonstration areas reached more than 80%,40%and 40%,respectively.
