Background and Objective:Studying pathophysiology mechanism of the chronic cervical spinal cord compression is the first way to cure the disease. The experiment uses self-made compression devices to press posterior cervical and thoracic cord, to build a model of chronic spinal cord compression, by which we can observe the behavior of the rats. We study the change of sense and motion of. four limbs, compare the number of apoptosis.We analyze the respective character of apoptosis at the different part of spinal cord.We test the expression of TNF-R1 and Cyto-c in the spinal cord, to research the mechanism of apoptosis.Methods:35 mice were divided into 6 weeks,12 weeks,18 weeks of compression group and control group, and 5 of normal contral. Every postanesthetic mice excises C5 and T5 laminae. Crop metal screws touch C5 and T9 dura mate of spinal cords. Gale combination score and BBB score are tested, and HE, TUNEL and TNF-R1 and Cyto-c staining are done. We compare apoptosis of nerve cells and analyze the mechanism of cell apoptosis. TTC staining compare area of ischemia and apoptosis.Rusults:1. Gale combination score and BBB score:The area of spinal canal is gradually reduced with the time going. The scores of 6 week's,12 week's and 18 week's compression group are clearly lower than those of control groups. With the increase of the period, the fall extent of score is gradually increasing.2. The shape of spinal cord:That of 6 week's shows a latent nick. That of 12 week's shows swelling and the shape of spinal cord is irregular,losing the former are contour. That of 18 week's shows that the compression area is clearly flatter and ellipse, especially cervical, with a deep nick in the middle.3. HE:6 week's shows irregular demyelination, but uniformity necrosis in the part of thoracic cord area.12 week's shows mass inflammation cellular proliferation around blood vessel in compression area, the gap and gliocyte hyperplasia significance.18 week's shows some apoptotic body. In thoracic cord, we can see new necrosis focus.4. TUNEL:The number of apoptosis cells in 6 week is little.12 week's mainly exists in white matter and grey matter junctional zone, the major part of which is gliocyte. We can see mass nerve cells apoptosis. in 18 week's compression group.5. TTC staining to observe the state of spinal cord ischemia:In all group, the whitening area is nearly the apoptosis area.6. immunohistochemisty staining:These posiyive cells reached to peak at 12 weeks.The former readued at 18weeks,but the later didn't.Conclusion:1. In the experiment, the area of spinal canal is gradually reduced with the time going, which imitates clinical spinal cord compression. The spinal column turns degeneration with the disease aggravation, which induce reduced area of spinal canal. Otherwise, we study the change of cervical and thoracic cord. We search the change at the different period and compression ratio, by taking sure that cervical and thoracic cord are made identical ratio compression.2. By scores, the animal spinal cord function is gradually reduced. The degree of reduction of oological behavioristics score is gradually increased with the compression time going, which means that animal's self-adaptation is decreasing with long period and compression.3. Compression makes the cervical cord flat, but by HE staining, the expression of thoracic cord is more serious than cervical cord. By TUNEL staining, apoptosis turns out in the thoracic cord after 12 week's compression. The time of apoptosis in cervical cord is late, but the number of apoptosis is more than that of thoracic cord, which means the cells in thoracic cord turns necrosis other than apoptosis with the compression time going.4. TTC positive staining area is close to that of apoptosis cell. TTC stainings in the 18 week's cervical and thoracic cord compression group are positive. The number of apoptosis cell in thoracic cord is obviously reduced.5. TNF makes apoptosis, to a certain degree, self -death style of spinal cord, and self-protection style of cells.the latter play a leading function at advanced stage because of the high expression of Cyto-c positive cells in long time.
