The effective material basis of Traditional Chinese Medicine(TCM)is the key to clarify the secrets of TCM and to enact the scientific quality control indicators.Based on the preliminary study and the overall concept of thinking of TCM with its characteristics of multi component and multi target integration,this topic research explored the correlation between effector substance and biological effect by using NMR-based metabonomics with multivariate statistical and bivariate correlation,aiming to reveal the effective material basis of Chinese medicine prescription Naodesheng(NDS).The completed studies are as follow:(1)Pharmacodynamic studies of 10 groups of NDS multiple combination samples on MCAO model rats were carried out by the experiments of neurobehavioral abnormality evaluation,TTC and HE stainings.They had varying degrees of effect on MCAO model rats,as evidenced by improved neurob ehavioral function,decreased cerebral infarct area and ameliorated pathological features,in particular,the effect of 4th,1st,3rd,6th,5th and 7th goups was significant.(2)Metabonomics studies of 10 groups of NDS multiple combination samples on MC AO model rats were carried out.They had varying degrees of effect on 6 biomarkers levels in model rat plasma,6 biomarkers levels in model rat urine and 12 biomarkers levels in model rat brain tissue,revealed by NMR-based metabonomics with PC A and PLS-DA.(3)Correlation study between effector substance and biological effect was carried out.3 9 components high relevant with biomarkers above were found by using bivariate analysis method,in particular,10 highest relevant components,including notoginsenoside R1,ginsenosi de Rgl,ginsenoside bl,ginsenoside Rd,hydroxysafflor yellow A,senkyunolide l,puerarin,daidzein,vitexin and ferulic acid.(4)The protective effect of components high relevant with biomarkers on model rats was determined.These 10 highest relevant components were found to improve neurobehavioral function,decrease cerebral infarct area,ameliorate pathological features and improve plasma LDH?MDA and SOD levels in model rats.Furthermore,these 10 components were found to reverse the biomarkers levels in model rat plasma,urine and brain tissue by NMR-based metabonomics.These results provided evidence for components of quality control for NDS.This topic research offers a feasible method and idea for the study on the effective material basis of TCM and components of quality control.
