| Objective: Based on the constitution theory of traditional Chinese medicine and the hypothesis of "long-term mental retardation,liver failure and accelerated brain aging",this paper,with the help of neuropsychological scale,biochemical detection method and neuroelectrophysiological technology,aims to find the early components of cognitive decline under the effect of age in people with qi stagnation constitution and peaceful constitution,to explore the influence of qi stagnation constitution on the aging of event-based prospective memory and its neuroelectrophysiological mechanism,and to provide theoretical basis and objective evidence for the early diagnosis,pathogenesis and treatment from the liver of patients with cognitive decline of qi stagnation constitution.Methods:From June 2019 to October 2020,the subjects were recruited from Zhengzhou,Zhumadian,Gongyi,Luoyang and other places in Henan Province.According to the inclusion and exclusion criteria,and according to the matching principle of age,gender and education level between groups,30 people in the qi stagnation constitution group and the control group were included in the TCM body mass scale in the 26-35 age group,36-45 age group,46-55 age group and 56-65 age group.In this study,the subjects of the same age group with different physique and the same physique with different age groups were respectively conducted a cross-sectional study of the overall cognitive function and cognitive domain evaluation,biochemical indicators detection,event-related potentials(ERPs)EEG information collection.(1)Montreal Cognitive Assessment(Mo CA)was used to detect the overall cognitive function and the differences in cognitive domains of qi stagnation constitution and peaceful constitution groups at different ages.(2)By enzyme linked immunosorbent assay(ELISA)to detect the differences of psychological stress(cortisol)and oxidative stress(peroxidase dismutase,malondialdehyde)between qi stagnation constitution people and peaceful constitution people under the effect of age,to explore the objective biochemical index basis of qi stagnation constitution affecting cognitive decline process,The correlation between oxidative stress and psychological stress was analyzed.(3)By using ERPs technology,we compared the average amplitude and latency of N100,P200 and P200,N300,PP components in the ongoing task and the prospective task of qi stagnation constitution groups and peaceful constitution groups in different age groups,and explored the differences of cognitive function between the subjects of the same constitution with different age groups and the same age group with different constitution.Results: 1.The main effect of age on Mo CA total score,visuospatial and executive ability score,naming score,language score,delayed recall score and abstract score was statistically significant(P < 0.05);There were significant differences in Mo CA total score,visual space and executive ability score,delayed recall score in the main effect of constitution(P< 0.05).Compared with the mild group,the score of qi stagnation constitution groups was lower than that of peaceful constitution groups;There was marginal significant difference between language score(P = 0.059)and abstract score(P = 0.073)in the main effect of constitution.The score of qi stagnation constitution was lower than that of peaceful constitution groups;In addition,there was a significant difference in the interaction effect between constitution and age(P < 0.05).The score of orientation in the 36-45 age group was significantly lower than that in the peaceful constitution groups.2.The main effects of age on serum cortisol,superoxide dismutase and malondialdehyde were statistically significant(P < 0.05).The levels of serum cortisol and malondialdehyde gradually increased with age,while superoxide dismutase gradually decreased with age;The differences of serum cortisol,superoxide dismutase and malondialdehyde in the main effect of constitution were statistically significant(P < 0.05).The levels of serum cortisol and malondialdehyde in qi stagnation constitution groups were significantly higher than those in peaceful constitution groups,and the level of superoxide dismutase in qi stagnation constitution groups were significantly lower than that in peaceful constitution groups.3.In terms of behavioral data,there were significant differences in the main effects of constitution and age on the correct rate and reaction time in both ongoing and prospective tasks(P < 0.05),The reaction time was prolonged.4.The ongoing task can induce N100 and P200 components in the anterior scalp(frontal area activation is obvious),and the prospective task can induce P200 components in the anterior scalp(frontal area activation is obvious),N300 components in the posterior scalp(occipital area activation is obvious),PP components(parietal area activation is obvious).Among them,except for the latency of N300 component of prospective task,there was marginal statistical significance in the main effect of lateralization(P = 0.068),the average amplitude and latency of N100 component of ongoing task,P200 component of prospective task and ongoing task,PP component of prospective task,and the average amplitude of N300 component,in the main effect of brain region and main effect of lateralization,The differences were statistically significant(P < 0.05).5.The difference in the main effect of N300 was statistically significant(P = 0.051).Secondly,there were significant differences in the latency of N100 in the ongoing task,N300 in the prospective task,and the average amplitude and latency of PP in the prospective task(P < 0.05).Moreover,with the increase of age,the average amplitude of each component has a trend of gradual decrease and the latency has a trend of gradual extension.6.In the main effect of constitution,the difference of P200 average amplitude in the ongoing task was statistically significant(P < 0.05),and the difference of P200 average amplitude in the prospective task was statistically significant(P = 0.053),that is,the average amplitude of qi stagnation constitution groups are more than that of peaceful constitution groups;In addition,there was a marginal statistical difference in the main effect of physical fitness in the latent period of N300(P = 0.085)and PP(P < 0.05),that is,the latent period of qi stagnation constitution groups are longer than that of peaceful constitution groups.Conclusion: 1.Qi stagnation constitution may be one of the risk factors of cognitive impairment similar to aging.The early manifestations of qi stagnation constitution on cognitive impairment may be mainly in executive function,delayed recall,language score,abstract score and orientation.2.Qi stagnation constitution may affect the aging process of cognitive function by intervening in the body’s psychological stress and oxidative stress system.3.Whether in the ongoing task or the prospective task,the aging trend of prospective memory was significant,and the change of behavioral response was the most obvious.N100,N300 and PP in ERPs may be the early sensitive indicators in the aging process of event-based prospective memory.4.In the aging process of event-based prospective memory,whether in the prospective task or in-process task,the change of cue awareness ability with aging is not obvious,and the transfer mechanism of cognitive processing components may be part of the mechanism.5.Compared with the people with peaceful constitution,the cognitive processing of event prospective memory in the people with qi stagnation constitution were relatively poor,and the behavioral change are the most obvious.In ERPs,the aging trend of qi stagnation constitution groups were more obvious than that of peaceful constitution groups in online retrieval and intention retrieval,which means that qi stagnation constitution may be responsible for the early damage of N300 and PP.6.The cognitive resources of cue awareness in qi stagnation constitution groups were more than those in peaceful constitution groups.This mechanism may be closely related to the Perfectionism of qi stagnation constitution group. |
