Background: Bladder cancer is the fourth most common malignant neoplasm among the world,with the morbidity of 7% and mortality of4%.Every year,more than 330000 newly diagnosed bladder tumors die of bladder cancer.Recurrence and metastasis are the characteristics of bladder cancer.It is of great significance to fully understand the molecular mechanism of bladder cancer.As a tumor suppressor gene,AT-rich interactive domain-containing protein1A(ARID1A),is often mutated or deleted in many types of tumors.However,its role in bladder cancer is still unclear.Objective: To investigate the expression of ARID1 A in bladder cancer and its relationship with prognosis,and to study the role and molecular mechanism of ARID1 A in bladder cancer.Methods: immunohistochemistry was used to verify the expression difference of ARID1 A in bladder tumors and normal tissues.The correlation between ARID1 A and clinical features such as pathological stages and grades was studied by ANOVA.At the same time,survival analysis was used to study the effect of ARID1 A on the prognosis of bladder tumors.We used siRNA to silence the expression of ARID1 A and study the effect of ARID1 A expression on the growth and proliferation of bladder tumor cells.Results: By immunohistochemistry,we found that the expression of ARID1 A was significantly different in bladder tumor and normal tissue,and it was lower in bladder tumor.Through analysis of variance,we found that the expression of ARID1 Awas significantly correlated with T stage and AJCC stage of bladder tumor;the results of survival analysis showed that the lower expression of ARID1 A significantly improved the overall survival rate and tumor specific survival rate of bladder tumor patients;And the ARID1 A is an independent prognostic risk factor for bladder cancer.In addition,patients with high expression of ARID1 A were more likely to benefit from postoperative adjuvant chemotherapy.Furthermore,the expression of ARID1 A was silenced by siRNA,the down-regulation of ARID1 A promotes the growth and proliferation of bladder cancer cells.In addition,the down-regulation of ARID1 A can also improve the migration and invasion of bladder cancer cells.Conclusion: The expression of ARID1 A is lower in bladder cancer;the expression of ARID1 A is significantly related to the stage of bladder cancer and the prognosis of patients;Patients with high expression of ARID1 A were more likely to benefit from postoperative adjuvant chemotherapy.The down-regulation of ARID1 A can promote the proliferation and migration as well as the invasion of bladder cancer cells.Therefore,our current analysis suggests that ARID1 A may be related to the occurrence and invasion of bladder cancer.Therefore,ARID1 A can be used as an independent diagnostic or prognostic biomarker for bladder cancer.
