With the nanometer-scale particle size and flexible controllability,the nano-drug delivery system overcomes the disadvantages of poor targeting,low bioavailability,great toxic and side effects,and single therapeutic effect in traditional treatments,and provides new possibilities the diagnosis and treatment of various major diseases.It is makes great sense to construct a multifunctional nano-drug delivery system for different types of diseases.Therefore,this article selects three different types of major diseases,tumors,Alzheimer’s disease and pneumonia,considering the types and pathogenesis of the diseases,construct three multifunctional and efficient nano-drug delivery system to achieve the diagnosis and diagnosis of these diseases.The details are as follows:1.Based on inorganic nanomaterials,aiming at the thermal intolerance characteristics of tumor cells and the structure of tumor tissues,we construct a NIR-stimulated combination therapy nano-drug delivery system:This chapter uses inorganic mesoporous silica nano-drug delivery system,loaded with liver cancer immunomodulatory drug sorafenib and photothermal diagnostic and therapeutic agent indocyanine green.It can retain in tumor tissue effectively and respond sensitively and accurately to the triggering of the 808nm laser.The drug delivery system can use fluorescence/photothermal imaging to monitor its distribution and metabolism in the tumor in a non-invasive manner,and can coordinate photothermal/immunotherapy,effectively killing tumor cells and increasing the secretion of IFN-γin cytotoxic CD8~+T cells,increasing the frequency of effect memory T cells,central memory T cells and NK cells in the tumor and spleen,enhancing the immune response and prevents tumor recurrence.This nano-drug delivery system has precise temporal and spatial control of the NIR,which minimizes adverse effects and has no obvious systemic toxicity.2.Based on organic nanomaterials,aiming at the abnormal accumulation of Aβand cholinergic imbalance in Alzheimer’s disease,we construct a serum albumin nano-drug delivery system for synergistic treatment,which loading the acetylcholinesterase inhibitor donepezil and metal ion chelating agent clioquinol.What’s more,the surface is modified with targeted peptides Aβtargeting molecule GM1 and penetrating peptide TAT.After nasal administration,the system can quickly reach the site of abnormal deposition of Aβin the brain,reduce the neurotoxicity induced by Aβand inhibit neuroinflammation in the brain;the release of the inflammatory environment and the inhibition of AChE further regulate the acetylcholine imbalance and delay the subsequent pathological cascade.dcHGT NPs has a protective effect on neuronal damage in the brains of AD mice,greatly alleviating the deficits in spatial learning and memory,and delaying the onset of AD.3.Based on organic nanomaterials,in response to the changes of the immune environment in pneumonia,we construct an integrated diagnosis and treatment serum albumin nano-Chinese medicine delivery system.Network pharmacology was used to screen the effective anti-inflammatory component trans-anethole in the traditional Chinese herb medicine fennel.Then we prepare the albumin nano-herb delivery system to co-loaded trans-anethole and indocyanine green.This system induces Treg differentiation and secretion of IL-10 cytokine in vivo and in vitro,inhibiting the M1 polarization of macrophages and the expression of TNF-α,and decreasing neutrophil infiltration to relieve the inflammatory storm in the lungs.TIB NPs have a greater ability to influence the immune balance and inflammation suppression.In summary,this article has designed and constructed a variety of multifunctional nano-drug delivery systems for different major diseases,which provides an experimental basis for the application and selection of nano-drug delivery systems in different disease models in the future.
