Soybean cyst nematode(Heterodera glycines Ichinohe,SCN),an important parasitic nematode on soybean,distributes in many countries(including China,America and Brazil)worldwide and causes huge losses to soybean production globally every year.In China,the 9 SCN races have been reported.Among them,SCN race 4 mainly distributes in Huanghuaihai soybean production area.It is well demonstrated that GmSHMT08 on Rhg4 and GmSNAP18 on rhg1 confer resistance to SCN.However,the mechanism underpinning the resistace still remains less well understood.In this study,a c DNA library was constructed using SCN race 4 and two bait vectors were constructed using GmSHMT08 and GmSNAP18 from soybean cultivar PI437654.Yeast two hybrid assays were performed to screen genes in SCN with interactive properties with GmSHMT08 and GmSNAP18.A gene encoding heat shock protein(HgHSP70)was selected as a candidate for interaction investigation.The full-length open reading frame of HgHSP70 was obtained by RACE(rapid amplification of c DNA ends)and then full-length coding sequence of HgHSP70 was cloned from SCN.Bioinfomatgics analysis revealed that most amio acids of HgHSP70 were hydrophilic and no transmembrane helices or signal peptide existed in HgHSP70.In situ hybridization revealed that HgHSP70 was specifically expressed in the intestinal cells.Exploring genes in pathogens that interact with host plant culd explain the the host plant’s resistance mechanism.In the research field concerning the interaction between host plant and nematode,most researches focused on the interaction between nematodes’ effectors and host plants,interaction between other proteins in nematode and host plants was less well characterized.This thesis discovered a solid interaction between HgHSP70 in SCN and soybean resistant gene GmSHMT08.This finding expanded our knowledge to the resistance mechanism of soybean against SCN,and can be used as a reference for the detailed study of GmSHMT08 medicated resistance against SCN.
