| The establishment of modern quality control and quality consistency evaluation method is the key to improve the quality of traditional Chinese medicine products.Although modern analytical techniques provide a powerful means for data acquisition of the quality control for traditional Chinese medicine,there are few studies on the methodology of quality control of traditional Chinese medicine,and the data processing methods are relatively simple and need to be improved.In order to solve the scientific problem of the quality evaluation of traditional Chinese medicine,this paper explores the quality control methods by regarding traditional Chinese medicine as complex material systems.By taking specific drug varieties as examples,new methods and new models of quality control and evaluation are probed by using multidimensional fingerprinting,chemometrics and informatics.1.The Linear Quantitative Profiling Method(LQPM)was established and expounded.By taking Alkaloids of Sophora flavescens(ASF)as an example,its quality was analyzed and evaluated by LQPM based on HPLC fingerprints.The fingerprint analysis was highly correlated to the quantitative analysis results of three marker compounds,and it is demonstrated that LQPM is able to obviously identify and discriminate the samples based on the difference in the quantitative content of all the chemical components.Furthermore,LQPM could employ the classical mathematical model to effectively quantify the multiple components of samples without any chemical standard by using composition similarities and related characteristic fingerprint.2.The connection method of HPLC fingerprints based on different separation mechanism was established,including signal connection method and data connection method.Taking ASF as an example,two HPLC fingerprints base on hydrophilic chromatography and ion suppression chromatographic separation mechanisms were established,respectively.By combined the ion suppression chromatography with mass spectrometry,the basic substances of ASF were studied.The fingerprints of the two separation mechanisms were processed by computer-aided signal connection and data connection,respectively.The results showed that the results of the signal connection fingerprint and the data connection method were accurate and consistent.The signal connection fingerprint can reveal the inherent characteristics of traditional Chinese medicine and it is better than single fingerprints in terms of signal intensity,separation degree homogeneity and information abundance.The data connection method can rapidly realize the comprehensive quality evaluation of the HPLC fingerprints under different separation mechanisms.3.The high performance capillary electrophoresis fingerprint combined with multicomponent quantitative analysis and antioxidant correlation was established as a model for controlling the quality of traditional Chinese medicine.Taking ASF as an example,the developed method was applied in developing capillary electrophoresis fingerprints coupled with quantitative determination of three components for assessing the quality consistency.The results show that the CEFP can effectively distinguish the quality of different samples,and provide a simple and feasible analysis method and strategy for the application of capillary electrophoresis in the consistency evaluation of traditional Chinese medicine.In combination with computational modeling study,it has demonstrated that significant differences in the complexation stabilities of the alkaloids with borate and citric acid is mainly responsible for the diastereomers separation of matrine and sophoridine.In addition,the fingerprint-efficacy relationship between chemical components and antioxidant activity in vitro was established using partial least squares regression model,which can serve as antioxidant activity evaluation of ASF as well as a reference for the selection of active constituents.4.The standard preparation control mode of traditional Chinese medicine was established.Taking Compound Liquorice Tablets as an example,HPLC quantitative fingerprints and UV spectroscopy quantitative fingerprints were respectively used to find the standard preparations by the systematic quantified fingerprint method.The two wavelength connection HPLC fingerprints can be used to maximumly reflect the fingerprint characteristics of the drug and avoid the one-sidedness of the single-wavelength evaluation.At the same time,the six active components can be measured.UVFP can monitor the overall characteristics of unsaturated chemical bonds,and provide a simple,high-throughput and low-cost new model for the selection of standard preparation.According to the selected standard of standard preparation,a total of 7 batches of samples were selected as standard preparation by integrating the HPLC and UVFP results.The mean fingerprints of standard preparation could be used as the standard fingerprints for recovering and correcting the standard preparation.The standard preparation control mode is the innovation of the quality control method of traditional Chinese medicine,which will provide new ideas and methods for evaluating the quality consistency of traditional Chinese medicine.5.A new intelligent model for predicting the quality of traditional Chinese medicine was established by merging HPLC fingerprints of crude drugs.Taking Compound Liquorice Tablets as an example,the HPLC fingerprints of licorice extract powder,powdered opium,star anise oil and sodium benzoate were merged to generate the intelligent mergence fingerprints.Its correlation with the raw materials and the compound preparation samples was also studied.The consistency of the intelligent merged fingerprints with the standard fingerprints was observed,and the results showed that the computer-aided model of intelligent mergence fingerprints could be used to predict the quality of traditional Chinese medicine compound preparation. |
