Bai Xuan Xia Ta Re Pian(BXXTR-FU)is a traditional Uighur medicine ancient prescription in China,which mainly consists of Euphorbiae Humifusae Herba,Chebulae Fructus,Terminalia Belliricae Fructus,Chebulae Fructus Immaturus,Aloe,Resina Scammoniae and other components.BXXTR-FU was widely used for the treatment of ringworm,tinea versicolor,psoriasis,atopic dermatitis,shingles,acne embolism etc.A large number of clinical practices showed that BXXTR-FU presented positive effect in the treatment of psoriasis,including not only a short course,but also high curative rate and few side effects.Based on the review of the chemical constituents and clinical application of BXXTR-FU,we preformed the microscopical identification of Euphorbiae Humifusae Herba,Chebulae Fructus,Terminalia Belliricae Fructus and Chebulae Fructus Immaturus,the characterization of chemical constituents,analysis of the absorbed components and the mechanism of anti-psoriatic lesions induced by Imiquimod(IMQ)in mice.The main chemical types of components in BXXTR-FU were analyzed by infrared three-stage identification technique.In FT-IR spectra,the double-peak 1616 and 1605 cm-1 of BXXTR-FU were considered as a sign of anthraquinones and their derivatives.In SD-IR spectra,the main carbonyl types of the absorption peaks of the BXXTR-FU in the range of 1743-1636 cm-1 were assigned:1651 cm-1 belonged to the carbonyl stretching vibration of flavonoids and chromones;1717 cm-1 belonged to tannins;1699 cm-1 belonged to carboxylic acid;1636 cm-1 belonged to anthraquinones and their derivatives.The correlations of the dynamic structural information among the chemical groups provided by 2D-IR spectra further confirmed the results of FT-IR and SD-IR spectra.Chebulae Fructus,Terminalia Belliricae Fructus and Chebulae Fructus Immaturus have similar plant species and chemical composition.In this study,a combination method of three-stage infrared spectroscopy and UPLC-TOF-MS technology was used to rapidly and accurately analyze and identify the aqueous extracts of them.A total of 57 chemical constituents were identified by using UPLC-QTOF-MS.BXXTR-FU could markedly alleviate IMQ-induced psoriasis by inhibiting IL-17-related inflammatory pathways.RNAseq analysis showed that BXXTR-FU significantly induced the expression levels of 8 genes.Additionally,12 absorbed components were determined by UPLC-QTOF-MS analysis,potentially targeted inflammatory related targets,which may be active constituents of BXXTR-FU in anti-psoriasis.The results indicated that BXXTR-FU could suppress psoriasis inflammation by multiple-constituents-regulated multiple targets synergistically.These findings shed light on the understanding of the active constituents and action mechanism of BXXTR-FU for the treatment of psoriasis and promoted the development of traditional Chinese medicine.
