Unsaturated fatty acids have many important physiological functions in organism. They are necessary for membrane structure and function. They also serve as precursors of eicosanoids, including prostaglandins and leukotrienes, and has a high antioxidant activity, an anti-cancer activity, and some other biological activities. Fatty acid desaturases are enzymes that introduce double bonds into the hydrocarbon chains of fatty acids to produce monounsaturated and polyunsaturated fatty acids. Some environmental factors (such as temperature, light, etc.) can impact the expression and activity of desaturases, which is achieved by some of the molecular mechanisms. The cells can quickly adapt to the changes in the environment by changing in the composition of fatty acids.Δ9 fatty acid desaturase is the first enzyme to catalyze the synthesis of unsaturated fatty acids, so there is great value for basic research. In addition, this paper also try to study the function of FAB2 from Chlamydomonas reinhardtii .This study was carried out by the following steps: First, The target gene FAB2 was cloned and its sequence was analysised through using some bioinformatics software. In addition, we also analysised the relevant characteristics of the predicted protein and its evolution which showed its higer homology with the gene from higher plants. The expression of FAB2 paves the way for the analysis of its bioactivities subsequently. The paper also studied the expression patterns of FAB2 under different stresses: first, we select several appropriate temperature and salinity gradients on the logarithm of the Chlamydomonas reinhardtii for induction and then analysis expression patterns of FAB2 by Real-time quantitative PCR. The relative expression of the gene was found increased to a certain extent at lower temperature and higher salty situations. Finaly, The function of FAB2 was researched. The method is firstly to build Synechococcus 7942ΔDesC mutants, and then put FAB2 gene into Synechococcus 7942ΔDesC mutants to identify the function of FAB2. The study of FAB2 from Chlamydomonas reinhardtii may show that temperature and salinity can impact the expression and activity of desaturases, which provide important clues for further studies of desaturases in other eukaryotes and also for the anti-stress study of plants.
