| In this study, research material is pig pancreas which are wasted by most slaughterhouse, pancreatin extracting was studied by the methods of isopropyl alcohol precipitation and chitosan precipitation to provide a reference for industrial production and process parameter. Activator and the protection of test were optimized; Through the single factor and the orthogonal test, the major effect factor and the process parameter of the pancreatin extracting method were studied, the main technical process and the parameter of pancreatin's product by two ways has determined, finally, the biological properties of pancreatin was studied. Through the pancreatin activities and physics chemistry Indicators were determined, the results fit for the chinese pharmacopoeia, the research results are as follows:1. Technical process: the methods of isopropyl alcohol precipitation and chitosan precipitation take the frozen pig pancreas as the raw material, during the pancreas broken, activation, extract, filter, precipitation, remove fat, vacuum drying to get the pancreatin.2. Through the single factor test, finally select after adding activator, than extract pancreatin with extracting solvents for best activation method, select the CaCl2—pancreatin for best activator combination. select starch, maltose as protective agent.3. Isopropyl alcohol precipitate pancreatin: Through the single factor test, determined concentration of Isopropyl alcohol, pH, extracting time, activation time, activation temperature, precipitation concentration as the main impact factors and then using L18(37)orthogonal test, take the activities of trypsin as the index, screened out the best process parameter were: extracting concentration 15%, pH 6.5, extracting time 3h, activation time 8h, activation temperature 8℃, precipitation concentration 60%. Result indicated that the yield of pancreatin was 10.79%, the activities of trypsin, amylopsin, lipase respective were 4791 U/g, 57806U/g, 45061 U/g, Loss on drying 4.61%, Fat content 18.19mg, the results fit for the chinese pharmacopoeia. 4. Chitosan precipitate pancreatin: Through the single factor test, determined concentration of alcohol, extracting time, pH, the amount of chitosan as the main impact factors and then using L9(34) orthogonal test, take the activities of trypsin as the index, screened out the best process parameter were: concentration of alcohol 10%, extracting time 6h, pH4, the amount of chitosan 0.1%. Result indicated that the yield of pancreatin was 9.24%, the activities of trypsin, amylopsin, lipase respective were 3703 U/g, 54285U/g, 44552 U/g, Loss on drying 4.86%, Fat content 17.58mg, the results fit for the chinese pharmacopoeia.5. Biological properties of pancreatin: The optimum temperature was measured 40℃, through the research of thermal stability, pancreatin activity decreased with the time gradually increased, the higher the temperature, the faster inactivation of trypsin. The optimum pH was measured 8, through the research of pH stability, pancreatin activity decreased with the pH gradually increased, the higher the pH, the faster inactivation of trypsin. According to research the affect of trypsin activity by Ca2+ and EDTA: Ca2+ does not affect the activity of trypsin, but EDTA has strong inhibition effect to the activity of trypsin.In summary, isopropyl alcohol precipitate pancreatin and chitosan precipitate pancreatin are both have feature as follows: Simple process, Short production cycle, high quality, but isopropyl alcohol precipitate pancreatin has higher yield, higher activity of three enzymes and best application prospect. So use advanced bioseparation engineering to produce pancreatin, increase the yield and enzyme activity which has positive significance in biochemical pharmaceutical industry and comprehensive utilization of animal by-products. |
