| Plants are subjected to varying degrees of environmental factors such as drought, extreme temperatures, salinity and other abiotic and biotic stress factors throughout their lives. Among these environmental factors, wind, rain, hail, touch, trauma, and the soil resistance, gravity, load, or negative pressure, the tension of plant growth, changes of cell turgor are all the mechanical stimulation. Mechanical stimulation on plant growth and development has a dual role. Low-intensity mechanical stimuli can promote the division and growth of plant cytoplasm, and high-intensity mechanical stimulation can influence the regular growth and development of plants, and even make plants die. However, how the plants recept and transmit the signal of mechanical stimulation is still unclear. In order to study the mechanism of plant response to mechanical stimulation, and the interaction between mechanical stimuli and brassinosteroid BR, we use molecular biology and genetics research for future study basis. We also constructed Artificial miRNA interference vectors.The major finding are as follows:(1)We use brassinolide synthesis defects of det2 mutants in the experiment. These three materials were treated by BR for 4 hours.Then,we use Realtime PCR to study the relationship between the regulation of the MSL gene family expression and the BR.The results show that gene expression of Arabidopsis MSL1-3 and MSL7-10 did not respond to the regulation of BR, MSL4-6 gene expression are up-regulated.(2)We use Western Blot method to detect MSL9 and MSL 10 protein expression under the control of BR.We found that MSL9-10 protein expression are down-regulated.(3)We cross the BR mutants bzrl-D withpMSL9-MSL9-GFP and pMSL10-MSL10-GFP transgenic plants.And we screened out bzrl-D×pMSL10-MSL10-GFP transgenic plants Successfully.(4)Artificial miRNA interference vectors are constructed by technology of Gateway successfully.These results suggest it may exist a relationship between mechanical stimulation and BR in plants, but also requires further study. |
