Ticks represent a group of highly specialized bloodsucking arthropods.They are well known vectors not only transmitting a great variety of infectious pathogens, but also having a super reproductive ability, and causing serious damage to human and domestic animals. At present, the reproductive physiology of ticks has become one of the international focus research realms. Vitellogenesis was the essential contents of this realms. Based on the purification of the vitellin(Vn), a series of researches were conducted on the vitellogenesis of Parthenogenetic populations of Haemaphysalis longicorni(sPPHL), including the physical and chemical properties of the vitellin and the vitellogenin. Many modern biological technologies and the methods of cross correlative realm, such as biological chemistry, analytical chemistry, GC/MS etc, were applied in these researches. The present work could be of important scientific significance in elucidating the vitellogenesis and regulation mechanism and revealing the reproductive biological laws, the adaptive strategy and the physiological regulative mechanism in ticks.The Vn was firstly purified from eggs of PPHL, which was only one kind of protein. The Black B and periodic acid-Shchiff (PAS) could stain Vn, and it revealed that Vn was a hemoglycolipoprotein. The isoelectric point of Vn was 5.4 and its relative molecular weight was 370 kD. Vn was composed of nine polypeptides and the relative molecular weight was 203, 147, 126, 82, 74, 70, 61, 47 and 31 kD respectively. The Vn lipids was detected by GC/MS, Lipids were mainly 2,4-di-tert-butylphenol, methyl 14-methyl pentadecanoate, Octadecanoic acid,methl ester, 9- Octadecanoic acid, methl ester. Lipids of ovary were detected by GC/MS in three stages including mating stage, the day of spawning and blood meal. The kind of lipid was similar but the percent of lipid was different slightly. There are eleven kinds of lipid, which were Phenol,2,4-bis(1,1-dimethylethyl), Pentadecanoic acid, 14- methyl- methyl ester, Benzenepropanoic acid, 3,5-bis(1,1-dimethylethyl) -4 -hydroxy- methyl ester, 9,12-Octadecadienoic acid, methyl ester, 9-Octadecenoic acid methyl ester,(E), Octadecanoic acid,methyl ester, Octadecanoic acid, 10-methyl-,methyl ester, 1-Cyclohexylnonene and Octadecenoic acid (Z)-,2-hydroxyethyl ester...
