Currently, 20 Arabidopsis glutamate receptor genes (AtGLRs) were discovered and divided into three subfamilies, whose amino acid coding sequences had high similarity with animal inotropic glrtamate(iGLRs) receptors. In animals, iGLRs had a function as glutamate- and glycine-gated non-selective cation channels(iGLRs), which played an important role in central nervous system. The funtions of AtGLRs were not very clearly, however, there were not directly evidence to prove their specific functions.In this study, though promoter fusion with GUS gene, 11 genes of AtGLRâ… a nd AtGLRâ…¢subfamilies were studied in terms of tissue localization. In addition, GFP fusion experiment was used to study the subcellular localization of AtGLR1.3, AtGLR3.1, AtGLR3.2, AtGLR3.3, AtGLR3.6 and AtGLR3.7 genes. The results showed that the expression of two subfamalies was partly organ- or tissue-specific. From the whole 10-day seedlings, the expression of AtGLRâ… was less than AtGLRâ…¢and even was not observed in root tip and hypocotyl, while the AtGLRâ…¢except AtGLR3.6 were highly expressed.However, the 11 genes expression also presenced overlapping phenomenon. Expression in stipule were observed from all genes, AtGLR3.5 and AtGLR3.7 could expresse in anther specifically, additionally, AtGLR3.2, AtGLR3.5 and AtGLR3.7 in developing seed. All genes expression showed the characteristic of temporal and spatial regulation. Generally, expression in young leaves was higher than old leaves, but AtGLR1.1 expression in old leaves trichomes was higher. Four gene expression would increase by implementing touch with old leaves. The protein of six genes were observed to locate in membrane by GFP fusion experiment.In order to study AtGLRs gene function advancely, AtGLRâ… and AtGLRâ…¢gene knockout mutants had been obtained from amiRNAs experiment.
