Gradute Xia XuechunAdvisor Professor Zhang YaozhouMany studies have suggested that modified hepatitis B surface antigensincluding preS sequences could be ideal candidates for new HBV vaccines withhigher efficacy. Several segments preS1(1-119aa),preS1(21-47aa) and S(1-223aa)were cloned into pBacPAK8 separately ane recombinant transfer vectorspBacPAKpreS1(1-119aa) and pBacPAKSS1(21-47aa) were got. Using linerizedbaculovirus expression system, recombinant viruses VpreS1(1-119aa andVSS1(21-47) were obtained, which were expressed in cultured Bm cells and larvaeof silkworm.The results of ELISA and Western revealed that the expression levels of therecombinant protein preS1(1-119aa) in Bm cells clould be 204ng/106cells, whichpresented at 14kDa and the recombinant protein SS1(21-47aa) reached223ng/106cells in Bm cells and 2.24μg/ml hemolymph in larvae of silkworms,whichpresented S and preS1 antigenicity and had two patterns at 27kDa and 29kDa.
