The In vitrO models of adipocyte differentiation were used to inveStigate theproliferation and differention of primary preadipocyte from rat adipose tissue in serumsuPPlemented.Preadipocytes were seeded in plating medium containing FBS--free(fetal bovineserum), l%FBS, l0%FBS and 20%FBS. PreadiPOcytes were seeded in diferent mediaincluding Ml99 medium, RPMIl640 medium and DMEM:HamF-l2(1:l) medium.Preadipocytes from adipose tissue of 20-d-old rat, 40-d-old rat and 60-d-old rat wereseeded in plating medium containing l0%FBS. Aftetr seeding by l6 hours, the numbersof preadipocytes dshed tO the platC were coamed by using a coulter counter. The rate ofadhension(RA) of l%FBS, l0%FBS and 20%FBS was higher than the RA ofFBS-free(P<0.0l). the RA of l0%FBS and 20%FBS was higher than the RA ofl%FBS(P<0.0l). the RA of l0%FBS was not different from the RA of 20%FBS. Theresults show that FBS have an significan effect on the attachInent of preadipocytes fromrat adipose tissue. the RAs of Ml99 medium, RPMIl640 medium andDMEM:Ham-Flz(l:l) medium were not different from each other. The attachInent ofpreadipocytes showed no dependence on media. The ax ofpreadipocytes from the adiposetissue of 20-d-old rat was more higher than that of 60-d-old rat(P<0.0l). the attchment ofthe preadipocytes from adipose tissue of young rat was more easily than that of old rat.FBS was added to the growh medium at concentration of l% FBS, 5%and l0%FBS. The proliferation of the preadipocytes of 5% FBS and l0% FBS were higher thanthat of l%FBS(P<0.0l). The proliferation of the preadipocytes of l0% FBS were higherthan that of 5%FBS(P<0.05). The exPeriment indicated that FBS have a highly signifinaneffect on the proliferation ofpreadipocytes. In order to describe the proliferating pattem ofpreadipocytes, the growth curve of the preadipocytes from adipose tissue of 20-d-old ratwas determined by the method of cell counting in l0%FBS supplemented.
