The purpose of this thesis is to explain, on the basis of the experimental data, the potential mechanism of monochromatic visible light's biostimulation on cells at a level which provides the scientific fundamentals of low intensity laser therapy. Low intensity laser therapy is a phototherapeutic method promoted within the last two decades as an effective tool for the treatment of soft tissue injuries, pain, arthritis and some other diseases. This technique is gaining widespread international acceptance among clinical professionals, but a certain disparity has arisen between the active work of physicians and the slow progress in laboratory studies. The present studies try to narrow this gap.The first chapter is devoted to investigate how He-Ne laser can induce respiratory burst of neutrophils. Here, we used some protein inhibitors to probe the signal transduction pathways involved in that process. Our results suggest that PTKs play a key role in the He-Ne laser-induced respiratory burst of neutrophils and [PTK-PLC-PKC-NADPH oxidase] signal transduction pathways may be involved in this process. As discussed in this chapter, we show a potential mechanism of low intensity laser biostimulationThe second chapter is devoted to a novel finding of LED irradiation on the cells. We found that LED irradiation (630 nm) decreased the Ap induced apoptosis of differentiated PC 12 cells. LED irradiation might increase the potential of mitochondria, while Ap 25-35 decreased it. The contradict processes may lead to that interesting result. As the Ap induced apoptosis plays an important role in the AD, LED irradiation, a promising tool, might be used to treat the AD.Chapter three is a theory study. Stem cell, with its special characters, might be affected by the low intensity laser or LED light. In this chapter, we discussed the possible biostimulation from the point of collective phototransduction and intracellular signal transduction.In other parts of the thesis, we have discussed some background of light biostimulation and the prospective using of visible light irradiation.
