The intestinal villus epithelia and microvascular are very important in digestion, absorption and mucosal barrier protection, through which all of the nourishment goes into the body. In order to study the changes of male rat jejunal epithelium's proliferation and apoptosis in different age and to study the morphological changes of other aspects, Sprague-Dawly rats were divided into four groups according to the development characteristic: natality (n=6), 3rd week postnatal (n=6), 3rd month postnatal (n=6), and 12th month postnatal (n=6). The rats were held for 12 hours without food. The mean weights were 5.6±0.3g for new born, 53.5±1.2g for 3rd week postnatal, 390?.9g for 3rd month postnatal and 601.7±3.4g for 12th month postnatal. Then the cell proliferation and apoptosis of the jejunal mucosal cells from natality to 12th month postnatal were observed with immunocytochemistry (ICC), TUNEL method. The height of jejunal villus, the thickness of jejunal muscle and the number of goblet cells of jejunal mucosal and glandulose epithelium were measured by BeiHang analytic software of morphometry and analyzed by STAT.The element of calcium is one of the necessary element which can maintain the physiological function, but when the absorbing of calcium was obstructed manifold diseases can be induced, like hypertension, hyperkinesias, colorectal carcinoma, cardiovascular disease and osteoporosis et al. Someone consider that the same cytoclasis mechanism which was arisen by outside factors is the lose contrast of balance in the cell. Many damaging factors like lack of oxygen, toxin, oxidative stress, defective blood perfusion, blood poisoning, ionizing radiation and enteritis et al. all can induce the consistency rising of the intracellularcalcium, which then result in the cell apoptosis. The interactional mechanism between calcium and iron is still ill-defined. The commonly thinking is that the ferric assimilating was hindered by calcium, but the reports on the influence of iron consistence change on calcium assimilating are only a few. So the influence of iron consistence change on calcium assimilating, the morphological change after the rising of calcium consistence and the apoptosis of Caco-2 cells were studied with confocal laser scanning microscope (CLSM) and Flow Cytometry (FCM) in Caco-2 cells, and then the elements and method of primary culture of intestinal epithelial cells (IEC) and the familiar problem and its overcoming measures were discussed, to offer the theoretical and morphological bases and the animal experimentation for basic and clinical study of digestive tract physiology and pathology, and also for aging biology.The research contents and results were as follows:1. The change of intestinal commonly structure with age(1)The height of jejunal villus was very small in natality (315.20±43.74), and then the jejunal villus were developed rapidly, the statistic result was 532.75 ±53.36 in 3rd week postnatal and was the highest in 3rd month postnatal (629.64 ±48.22), and the villus was swelled and shortened in 12th month postnatal (510.95 + 82.93). The height of jejunal villus in 3rd week postnatal was obviously more than that of natality (.P<0.01), and the height of jejunal villus in 12th month postnatal was obviously less than that of 3rd week postnatal (P<0.01) analyzed by STAT, which can be the cue on that the height of rat intestinal villus was heighten with the enhancing of its assimilating function, and so the sufficient surface area was provided for the intestinal assimilating function. When the rat was aged the intestinal villus appeared the caducity change of shortened and swelled, by which the assimilating area was shrinking, and this change was the main factor of inducing the gradually decrease of intestinal assimilating function. (2)The thickness of the jejunal muscle layer was taken onincreasing current from natality (32.0±15.71) to 3rd week postnatal (64.86± 14.91), and the muscle fibres of rat jejunal muscle layer was very loosen in 3r week postnatal.
