The family of AP-2 proteins is a very important family transcription factors. Four members of the family have been cloned , which are AP-2α, AP-2β , AP-2γ , AP-2δ. In this study, the AP-2β gene was fused to DNA binding domain of GAL4 in the expression vector PDB-Leu of the yeast two hybrid system . The clones were sequenced to confirm the reading frame. The correct clone was then used to transform the yeast Mav203. Two positive clones that interact with AP-2β were identified in a yeast two-hybrid system screen of a heLa cDNA library. The interaction was further confirmed by Colony lift filter assay . Sequencing result indicates that these two clones are CKII and GAS41.
