In the study, isolate a thermophiles strain desigated TTH from 80℃ hot spring at xiji village, He-Zhou county in Guangxi. Gram-nagative, coloy was round,smooth on the surface, the cells were rod-shape, without flagellum and spore. Strain TTH grew at temperatures between 55 and 85℃, with an optimum at between 70 and 75℃. pH was detected between 5.0 and 9.0, with an optimum at 7.0 and 7.5. Salt concentration for growth was around 0 to 1.2%(wt/vol), the optimal concentration was from 0% to 0.4%. TTH was strictly aerobic. The G-C content was 65.6 mol%. It belong to Thermus genus by 16S rDNA sequence comparison. The strain was highly related to Thermus thermophilus CS, Thermus thermophilus HB8 and Thermus thermophilus X6. Their simility were 98.6%, 98.5%, 98.5% respectively .The phylogenetic tree showed that the new isolation was different from three strains. It was proposed to represent a novel strain within the Thermus thermophilus. Isolation and identification of TTH made us to recognize deeply microbial resource of the hot spring in Guangxi.Strain TTH could grew at the medium with strepcomytic, kanamyicin and tetracycline, it was inhibited by ampicillin. Acording to the characteristic, try to extract its plasmid, the result showed that strain TTH contained one plasmid , transformed the plasmid into E.coli, E.coli can grew at LB medium with kanamyicin, extract the plasmid from E.coli, the size was the same to strain TTH. The result of sequence showed that pTTH was homology to Thermus thermophilus HB27 plasmid pTT27. It was not reported in the international.
