Study on structural analysis and biological activities of polysaccharides from fermentation filtrate of Tremella aurantialbaYun-Xia DengSupervisor: Professor Wei-jing Qu Major:BotanySubject:Source Plant ChemistryIn the first place, the extro-polysaccharides from Tremella aurantialba (TEP) was isolated, purified and primarily studied on its structure. TEP include polysaccharide 51.8%, sulfate radical 0.64%, uronic acid 12.8% by chemical determination. Its mono-saccharide composition was identified by thin-layer chromatography and HPLC.TEP consists of rhamnose, arabinose, xylose, fucose, glucose, galactose. Component a and component b of polysaccharides were isolated and purified by column chromatography on DEAE-sephadex A-25 and Sephadex G-200 from TEP. Their structures were primarily studied by TLC, periodic acid oxidation and Smith's degradation, IR and so on. The results showed that both of them consists of rhamnose, arabinose, xylose, fucose, glucose, galactose (Component a and component b), and there existed glucosidic bond, there were there lingking ways:-(1→3), (1→4)及(1→6).In order to making clear the physiological function of TEP, TEP was investigated in normal mice and diabetic rats induced by alloxan and STZ respectively. 100, 200, 300mg/kg.d of TEP were taken orally by normal mice, The result showed that TEP have no effect on the level of plasma glucose in normal mice;but 200,300mg/kg.d doses of TEP can significantly reduce the level of plasma cholesterol compared with the control(p<0.05) for 3w. And the gluconeogensis can be inhibited obviously compared with control.After given TEP 21 days, the death rate of mice induced by STZ weresignificantly reduced.Effect of TEP on STZ-inducing diabetes mellitus model was investigated by applying doses(150mg/kg.d), compared with the control, the level of plasma glucose reduced, but didn't have the remarked difference(p>0.05), and the same as the level of serum cholsterol, triglyceride, fruc level and weights in diabetic rats.Effect of TEP on alloxan-inducing diabetes mellitus model was investigated too, we received the same results.For making clear the mechanism of the results, we make in-depth studies.The experiments of lipid peroxidation induced by Fe2+-ascorbic acid in mouse liver microson and the hemolysis of rat RBC induced by H2O2 showed the TEP have distinctive effect of antioxidation.To investigate inhibitory effect of TEP on the nonenzymatic glycosylation of Protein in vitro, we found its inhibitory rate is 27% lower than that of AG, MFH. And TEP can also enhance the clearance rate of iv charcoal particles in mice (p<0.05).
