y -Linolenic acid(GLA) is a polyunsaturated fatty acid (PUFA) With important physiological functions to human body. Compared with the method of chemical synthesis and extracting from plant seeds and animal fats, microbial fermentation has its unique advantage, therefore, it is necessary to breed a high-yield strain and to find out its optimum fermentation conditions.In this article, numerous protoplasts of strain C.echinulata Thanter Rl was produced in shaking flask and higher recurrent ration was gotten in two lays cultivation. Then, Mutant strain M2040 accumulating a large amount of lipid was successfully obtained when the protoplasts or spores of original strain Rl was treated by He-Ne laser and LiCl as single or compound mutagen. The content of lipid, the production of lipid, the production of GLA in M2040 cultivated in shaking flask was 146.2%, 96.5%, 27.6% and 209.1% higher than that in the original strain Rl despectively. when cultivated in liquid state fermentation, M2040 and Rl were analyzed and identified by morphological character, Infrared absorption and the lipid producing ability. The results proved the genetic materials of Rl had already changed after the mutation. After times of transplantation, the content of lipid maintained showed that the genetic characteristics of mutant M2040 was stable.After studies on lipid producing conditions of M2040 by different single factor, the optimum fermentation composition and conditions for lipid production are gotten, on the ground of which we put up the two stages of fermentation, then,the production of mycelium, lipid and GLA in M2040 cultivated in shaking flask was 122.9%, 195.2% and 281.8% higher than that in the original strain despectively.
