EXP.1: Collectivity distribution and decrepitude change of NOS positive neurons in rabbit brain,were studied with NADPH-disphorase histochemical method.The results showed as follows:(â… ) NADPH-d positive neurons distributed in wide areas of rabbit brain.They include cerebellum,cerebral cortex,hypothalamus, mesencephalon, and pons.A few of positive cells could be seen in the medulla oblongata.(â…¡) NADPH-d positive neurons appears blue.Their shapes were diversform such as polygon,fusiform,nummular and oval etc. Karyon hasn't pigmentation. Dendriate and axon's pigmentation are clear.(â…¢) This experimentation results showed that NADPH-d positive neurons go through such course as the following:With the growth of rabbit NADPH-d positive neurons gradually largen, its shape gradually mature.But when the rabbit turns effete,its density significantly decreases. Comparing the adult rabbit with the young, the juvenile, the youth rabbit,the diameter and shape of the tuber haven't significant change.But comparing the aged rabbit with the adult rabbit,there was a significant change. The results indicate that as a neurotransmitter NO is probably related to the development of the neuron.EXP.2: Immunocytochemical SABC were conducted to detect and determine the distribution and decrepitude change of nNOS immunopositive neurons in rabbit brains. The results showed as follows: (â… ) nNOS immunopositive neuron distributed in wide areas of rabbit brains.They include cerebral cortex,cerebellum, mesencephalon, pons and hypothalamus.A few of nNOS immunopositive cells could be seen in the medulla oblongata. (â…¡)nNOS immunopositive neurons were brown and their pigmentation was maily located in cytoplast,the pigmentation was very light in karyon.Their shapes were diversiform such as triangle spherical, nummular, elliptic and fusiform etc.They had one or several tuber.The immunopositive fibers were strings of pearls,and crossed each other. (â…¢) This experimentation results showed that development level of nNOS immunopositive neurons approach the adult rabbit when it is a young rabbit. The effete change appears when rabbits turn ageing.That was expression intensity weaken. The results: The extensive distribution of nNOS immunopositive neurons indicates that NO is involved in many functions of CNS,such as regulating domination in nervous system and the development of neuron.EXP.3: Immunocytochemical SABC were conducted to detect and determine the distribution and decrepitude change of eNOS immunopositive neurons in rabbit brains. The results showed as follows: (â… ) eNOS immunopositive neurons and fibers were distributed in extensive region of rabbit brain,including diencephalon,cerebral cortex and cerebellum.but eNOS were almost negative in brain stem. (â…¡) eNOS immunopositive neurons were brown and immunopositive matter was maily located in cytoplast in which pigmentation was uniformity, while the pigmentation was very light in the karyon.Their shapes were diversiform such as nummular, elliptic and oval etc. Positive neurons had one or several tubers and its axon was short. (â…¢) This experimentation results showed that development level of eNOS immunopositive neurons approach the adult rabbit when it is a young rabbit. The effete change appears when rabbits turn ageing.That is expression intensity weaken.EXP.4: This experiment was to investigate the changes of NOS activities in rabbit brains along with the increase of the rabbit age.Method: Biochemistry reagent kit determination technology.The results showed:the NOS activity in cerebellum was highest in rabbit brain, secondly was diencephalon,thirdly was brain stem, cerebral cortex was the lowest.The NOS activities in rabbit brain are depressed with aging,but there distinguishing differences in regions.
