In this study, Fluorescence in situ hybridization (FISH) was successfully applied in the chromosome analysis and gene mapping of Fenneropenaeus chinensis and Ciona?intestinalis by the successive chromosome preparation, probes labeling, and hybridization. This study will facilitate mapping of gene, constructing of physical map and evolution research in Fenneropenaeus chinensis, Ciona?intestinalis and other invertebrate.Chromosomes with high quality were obtained from the embryo of Fenneropenaeus chinensis and Ciona?intestinalis. Analysis indicated that the karyotype of Fenneropenaeus chinensis was: 2N=88=62M+16SM+6ST+2T, NF=174, and the karyotype of Ciona?intestinalis was 2N=28=18M+6SM+2ST+2T,NF=54. The smallest chromosome of F. chinensis, which was identified as spot-like chromosome before, was proved to be telocentric in this study.In addition, two conservative genes were localized on the chromosomes of F. chinensis and C.?intestinalis. hsp70 was localized at three pairs of chromosomes of F. chinens with one locus on each chromosome. Actin gene was localized at two pairs of chromosomes with 3 loci on each chromosome. In C.?intestinalis, actin gene was localized on one pair of homologous chromosomes with one locus on each chromosome.
