Cytoplasmic polyhedrosis viruses(CPVs)belong to the genus Cypovirus in thefamily Reoviridae.Their host range is restricted to Lepidoptera,Hymenoptera,Dipteraand Coleoptera.So far ,it has been found that CPV infected about 200 speciesinsects.These viruses produce protein occlusion bodies called polyhedra in thecytoplasms of infected midgut epithelial cells of insects.The cypovirus genomeconsists of ten segmnent of double-stranded RNA (dsRNA).Lack of appropriate cellculture systems,together with the high levels of diversity that exist between differentcypovirus species,have restricted knowledge concerning the exact number andfunctions of the proteins of any individual cypovirus. In the paper,the research of the clone and Prokaryotic Expression of p44 andp119 gene of Dendrolimus punctatus cypovirus WenShan strain have been done.p44gene was cloned to expression vector pET-28a and induced by IPTG.the condition ofinducing was optimized.The p44 gene has 1174 bp which encode 390 amion acid withmolecular masses of approximately 43 kDa(p44).Sequence analysis of the deducedamino acid sequence of DpCPV P44 revealed possible sequence homology withprotein from BmCPV S8,LdCPV-1 S8,Mutase.This may suggest that P44 isnon-structure protein.And p119 gene was cloned and sequenced,it contains3177-nucleotide sequence which is predicted to encode a protein with a molecularmass of about 120kDa.sequence analysis of the deduced amino acid sequence ofDpCPV P119 revealed possible sequence homology with protein rice ragged stuntvirus(RRSV)S2 and Rice bunchy stunt virus(RBSV)S3.
