| Artemisia sphaerocephala Krasch. is widely distribute in Gansu and Neimenggu Province, China. The surface of its seeds is covered by a layer of gum, Artemisia seed polysaccharide (ASP). ASP has been found to possess high viscosity which is 1800 times as much as that of gelatin and strong hydroscopicity, for which it can absorb 60 times its self weight of water, and there are aslo a great of oil in its seeds, Artemisia oil(AO). The content of linolic acid is more high in AO. So, it is important to developing the seeds of Artemisia sphaerocephala Krasch.1. In this paper, ASP was extracted with hot water.The best conditions were determined by orthogonal test. Respectly, the temperature, time, the ration of solvant and solute is 70℃,1h, 1:150.2. The sevage method and papain was used together in deprotein, which is appropriate way. The physical characteristic was measured with advanced instrument. The weight of molecule of ASP is 1.42 × 105g/mol. The specific rotatory power is +90° . The total content is 96.5%. And ASP is reducibility polysaccharide.3. ASP was prepared as acetate derivatives. The products were analysed by GC and GC-MS. The result shows ASP is composed of L-Ara, D-Xyl, D-Lyx, D-Man, D-Glu, D-Gal. The molar ration is about 0. 14:0. 09:1. 51:0. 17:0. 07:1. 45. The structures is characterized with FT-IR, 1HNMR, 13CNMR. The results reveal that the monosaccharide components have α -and β -, but β - is the main way. ASP may have following linking-way. the main linking way of Glc is (1→), there were structural units between Glc and Glc with β -1,4. In the meantime, the main linking way of Man is also (1→); and have linking way of Man(l,3).There are aslo structural units between Man and Man with α -D-1, 6. In addition, Gal have a -or β - and 1 → or 4→.4. The images was studied by AFM.The results demostrate ASP is linear molecule with branches. The diameter of single molecule is less than 2.53nm. It can be formed the network structure by the entanglment between the single molecules.5. The Ao was obtained by CO2 supercritical method. The fatty acids of AO was analysed by GC. There are twelve fatty acids in it. The content of linolic acid is 56.02%. |
