To get healthy and long-life sheep follicular granular cell (FGC) lines, we design the following experiment. This experiment include four parts:1 .Establish a method of culturing normal sheep follicular granular cells (FGCs) by study various culturing methods.2. Introduce plasmid pLPC-hTERT into FGCs, and get a positive FGCs by puromycin selection.3. Chromosome analysis checkes the possible of malignant transfomation.4.RT-PCR, TRAP-silver staining and southern are used to analyze the telomerase activity and the telomere maintenance.The results of the experiment show the positive FGCs live longer than normal follicular granular cells, and the sign of malignant transformation have not been found. Meanwhile, the molecular analyses are positive in the transformed FGCs. At last, we get the healthy and long-life FGCs, and this can provide a stable somatic cell line for gene target.
