In this paper , the author studied the secretion of exopolysacchride of Chlamydomonas sajao Lewin under different culture conditions especially under the different concentrations of NaHCO3, Ca(N03)2, and KH2PO4. We find that with the decrease of the NaHCO3, the produce of the inner polysaccharide decreased while extra polysaccharide increased, which means that the low concentration of N'aHCOi improves the secretion of the extra polysaccharide. Different concentrations of Ca(N03)2 don't make much difference. When there is short of KH2PO4, the secretion of the exopolysacchride enhanced remarkably.Separated and purified the extra polysaccharide of shajiao , The crude exopolysacchrides were isolated from the cell culture of shajiao after the medium being centrifugated, evaporated, dialyzed and precipitated with ethanol. The pure extra polysaccharide was obtained by purification on DEAE-cellulose columns. It was identified by Sephadex G-200 gelatin column chromatography to be homogenous. Chemical analysis showed that the polymer contained uronic acid and sulfated groups, which account for 12. 5%, 2. 08% of the pure extra polysaccharide weight.With the increase of the concentration of extra polysacchride, the soil water-holding capacity is increase obviously. The high concentration of exopolysacchride in soil retards the water movement through the soil and retard the soil water-evaporation rate too.We discussed the relation between the Phosphorus density of medium and the alkaline phosphatase activity of the Algae-Rice (O. sativa L ) Experiment System. The authors design three culture mediums: having phosphorus, without phosphorus and having phosphate rocks. The authors find out that alkaline phosphatase activity was increased when Chlamydomonas sajao Lewin was transforming and availing the phosphate rocks. Alkaline phosphatase is a very important isozyme taking parting in Phosphorus-metabolism. Therefore, the authors consider the variation of alkaline phosphatase activity is a symbol that the algae accommodate itself physiological condition to adapt to transform and avail phosphate rocks.
